| Objective:To explore the expression relationship of Hoxall, Integrin-β3, β-catenin, leukemia inhibitory factor (Lif) on the gene and protein level, up-regulate and down-regulate the expression of Hoxall in mice uteri through transfected with plasmid-liposome complexes.Methods:Select nulliparous sexual maturity Kunming mice divided into two groups (Using self-control):The first group were injected the pEGFP-Hoxall and empty plasmid-liposome complexes into both sides of Y-shaped uterus of pregnancy D2mice respectively; the other group were injected with Hoxall SiRNA and empty ones. Excise the uteri tissue after the mice euthanized by cervical vertebrae dislocation under anesthesia on Day5. Then screen out the mice of the expression of Hoxall up-regulating and down-regulating successfully through Realtime PCR and Western blot on the levels of mRNA and protein, at last detected the expression of Integrin-β3, β-catenin and Lif.Results:Levels of Integrin-β3, β-catenin and Lif mRNA and protein were dramatically increased and decreased in endometrium transfected successfully with pEGFP-Hoxall and Hoxall SiRNA compared with the corresponding control ones, consistent with the expression of Hoxall.Conclusion:The expression of the four factors may influence each other in micro-environment of endometrium. Hoxall may influence endomtrial receptivity through altering the expression of endometrial integrin-β3,β-catenin, and Lif. Objective:To research the relationship of expression of Hoxall, Integrin-β3, β-catenin, and Lif in the peri-implantation endometrium in infertility women with endometriosis by Realtime PCR and Western blot, further to explore the mechanism of the four factors regulating endometrial receptivity.Methods:Detect the expression of Hoxall, Integrin-β3, β-catenin, and Lif in endometrial tissues from16infertile patients with endometriosis (endometriosis group) and20control women with tubal obstruction (tubal obstruction group) in peri-implantation endometrium through Realtime PCR and Western blot, compare and analysis the relations between them, then follow up the pregnancy outcome of the two groups of patients with infertility ongoing IVF-ET, further evaluate the importance of the four factors in endometrial receptivity.Results:Levels of Hoxall, Integrin-β3, β-catenin, and Lif mRNA and protein were significantly decreased in endometrium of endometriosis group compared with the tubal obstruction group, and the implantation rate and clinical pregnancy rate in endometriosis group are much lower than the tubal obstruction group after IVF-ET.Conclusion:Impaired down-regulation expression of Hoxall, Integrin-β3, β-catenin, and Lif might be one of the potential molecular mechanisms of infertility in patients with endometriosis. They play a role in implantation process and coordinate with each other. |
