Experimental study on dose-effect and time-effect of the combination of Sophora flavescens and schisandra on improving sleepPurpose: The threshold dose is selected to lay the foundation for the further joint use of synergists(such as 5-HTP)and antagonists(such as PCPA).Materials and methods:1.Experiments on the effects of Sophora flavescens and schisandra on the sleep of mice induced by pentobarbital sodium,Carry out dose-effect tests and time-effect tests,Determine the effective dose of the drug and the administration cycle.2.The sleep incubation period and duration of sleep in mice were investigated.Results:1.The experimental results of dose-effect relationship show,the compatibility of Sophora Flavescens and Schisandra chinensis is 1.04 g/kg,2.08 g/kg,4.16 g/kg,8.32 g/kg,the effects of sodium pentobarbital on sleep in mouse were significantly reduced(P < 0.01);Extended sleep duration(P < 0.05,P < 0.01,P < 0.01,P < 0.01),there was a dose-response relationship.2.Experimental results of time-effect relationship show,dosing one day,three days,five days,seven days,there were no significant differences in sleep latency and duration of sleep between the blank groups(P > 0.05).One day after dosing,compared with the blank group,the Sophora flavescens and Schisandra chinensis group(1.04 g/kg)has a tendency to shorten the sleep incubation period and prolong continuous sleep time,but there was no significant difference(P > 0.05).For three consecutive days,compared with the blank group,Sophora flavescens and Schisandra chinensis groups(1.04 g/kg)had the effect of shortening the incubation latency(P < 0.05),there was a tendency to prolong the duration of sleep,but there was no significant difference(P > 0.05).For five consecutive days,compared with the blank group,Sophora flavescens and Schisandra chinensis group(1.04 g/kg)had the effect of shortening the incubation period(P < 0.05),it has the effect of prolonging the duration of sleep(P < 0.01).For seven consecutive days,compared with the blank group,the Sophora flavescens and Schisandra chinensis group(1.04 g/kg)significantly shortened the incubation latency(P < 0.01),it has a significant effect on prolonging the duration of sleep(P < 0.01).Conclusion: The effective dose of Sophora flavescens and Schisandra chinensis to improve insomnia is 1.04 g/kg,and the administration period is seven days.Experimental study on the Sophora flavescens of matrine and schisandrae in improving sleepPurpose: Clarify the target and mechanism of the drug to improve sleep,and lay the foundation for the rational clinical application of the drug.Materials and methods:1.Effect of Sophora flavescens and Schisandra chinensis combined with flumazenil on pentobarbital sodium-induced sleep in mice.2.Effects of Sophora flavescens and Schisandra chinensis combined with 5-HTP on pentobarbital sodium-induced sleep in mice.3.Effect of Sophora flavescens and Schisandra chinensis combined with PCPA on pentobarbital sodium-induced sleep in mice.4.Enzyme-linked immunoassay was used to investigate the effect of Sophora flavescens and Schisandra chinensis on the content of substance P in the brain.5.Enzyme-linked immunoassay was used to investigate the effects of Sophora flavescens and Schisandra chinensis on the content of IL-1? and TNF-? in the whole brain of mice.Results:1.The effect of Sophora flavescens and Schisandra chinensis combined with flumazenilon the sleep of mouse induced by sodium pentobarbital,compared with the blank group,each group,there are significant differences in sleep latency and duration of sleep,thisindicates that the pentobarbital sodium induced a successful replication of the mouse sleep model.Compared to the model group,the flumazenil group reduced sleep time,all other groups significantly prolonged the sleep duration of mouse caused by sodium pentobarbital(P <0.01),compared with the flumazenil group,each administration group significantly prolonged the sleep duration of mouse caused by sodium pentobarbital(P < 0.01);The sleep time of Sophora flavescens and Schisandra chinensis group and Sophora schisandrae and Flumazenil group significantly prolonged sleep time(P < 0.05).2.Experiments based on 5-HTP energy nervous system-mediated bitter participation of Schisandra chinensis to improve sleep,compared with the blank group,There are significant differences in sleep latency and duration of sleep,this indicates that the pentobarbitasodium induced a successful replication of the mouse sleep model.Compared to the model group,Each group significantly prolonged the sleep duration and shortened the sleep latency of mouse induced by sodium pentobarbital(P < 0.05,P < 0.01),Compared with the 5-HTP group,each administration group could significantly prolong the sleep duration and shorten the sleep latency of mouse induced by pentobarbital sodium(P < 0.05,P < 0.01).And has synergy with 5-HTP,sleep latency was shortened(P < 0.05 or P < 0.01)and sleep duration was prolonged(P < 0.05 or P < 0.01).3.Effect of Sophora flavescens and Schisandra chinensis combined with PCPA on the sleep of mouse induced by sodium pentobarbital.Compared with the blank group,there are significant differences in sleep latency and duration of sleep,this indicates that the pentobarbital sodium induced a successful replication of the mouse sleep model.Compared with the model group,the PCPA group has the effect of shortening sleep time,the other groups have the effect of prolonging the sleep duration and shortening the sleep latency of mouse caused by pentobarbital sodium(P < 0.05,P < 0.01).Compared with the PCPA group,the sleep latency of each group was significantly shortened,and the sleep latency of the Sophora flavescens and Schisandra group was shortened and the sleep time was extended.In addition,compared with the PCPA group,the dosing group significantly reduced the sleep latency and prolonged the duration of sleep(P < 0.01,P < 0.01).4.Test of improving sleep effect based on compatibility of Sophora flavescens and Schisandra chinensis based on peptides,The combination of Sophora flavescens and Schisandra chinensis in high,medium and low dose groups can significantly reduce the content of substance P in the whole brain of mouse,and the difference is statistically significant compared with the blank control group(P < 0.05 or P < 0.01).5.Test of improving sleep effect based on cytokine-mediated compatibility of Sophora flavescens and Schisandra chinensis,The combination of Sophora flavescens and Schisandra chinensis in high,medium and low dose groups can significantly reduce the IL-1? content in the whole brain of mouse,compared with the blank control group,the difference was statistically significant(P < 0.01).The combination of Sophora flavescens and Schisandra chinensis in high and low dose groups can reduce TNF-? content in the whole brain of mouse,There was a significant difference between the high and low dose groups of Sophora flavescens and Schisandra chinensis and the control group(P < 0.05).Both the high and low dose groups of Sophora flavescens and Schisandra chinensis can reduce the TNF-? content in the whole brain of mouse,and the difference between the high and low dose groups and the blank control group is statistically significant(P < 0.05).Conclusion:1.The combination of Sophora flavescens and Schisandra chinensis has an antagonistic effect on flumazenil;2.Compatibility of Sophora flavescens and Schisandra chinensis with 5-HTP;3.Compatibility of Sophora flavescens and Schisandra chinensis on PCPA;4.Compatibility of Sophora flavescens and Schisandra can reduce the content of substance P in the whole brain of mice;5.Compatibility of Sophora flavescens and Schisandra can reduce the content of IL-1?
