Currently,lung cancer has become the leading cause of cancer death worldwide.Nonsmall cell lung cancer,as a typical subtype of lung cancer,accounts for more than 86% of the total number of lung cancers and is one of the most common malignant tumors.Chemotherapy,surgical resection,radiotherapy and some other therapies are currently the main strategies for the treatment of non-small cell lung cancer.The traditional Chinese medicine Sophora flavescens is the root of Sophora flavescens,which has been used clinically to treat a variety of diseases,but its underlying mechanism of action remains unclear.In this study,we used an innovative systemic pharmacology platform to systematically and multi-levelly reveal the pharmacological mechanisms of Sophora flavescens in the treatment of non-small cell lung cancer.First,we applied the ADME/T assessment to screen out the active constituents in Sophora flavescens,and predicted the corresponding targets and pathways of these components,and studied the mechanism of the efficacy of Sophora flavescens from the molecular level,target level and pathway layer.The corresponding molecular-target map and target-pass map were constructed.The results showed that 45 active compounds in Sophora flavescens have potential anti-tumor effects.The corresponding 157 targets of these compounds involve many biological processes such as angiogenesis,apoptosis and inflammation.Animal experiments combined with in vitro experiments such as cell viability assay,Western blot analysis,real-time quantitative PCR,etc.,confirmed the reliability and accuracy of the predicted data.In conclusion,this study demonstrates that Sophora flavescens can treat non-small cell lung cancer to a certain extent,and has proved its mechanism of action,providing theoretical support for the anti-cancer of Chinese medicine,and promoting the development and application of traditional Chinese medicine.
