The alkaloids and flavonoids are one of the important components in Sophora flavescens Ait. However, people had been put the high emphasis on alkaloids in the past. So the content of alkaloids was only determined in Sophora flavescens Ait. For appreciating the quality of Sophora flavescens Ait. Obviously, it's difficut to reflect the quality of Sophora flavescens Ait. On the development of the study on Sophora flavescens Ait., these pharmacological actions of the flavonoids in Sophora flavescens Ait. have been shown , such as anti-inflammation, anti-bacteria, anti-virus and so on. It's very important for studying and exploiting the flavonoids in Sophora flavescens Ait. Pass the extraction and separation of the flavonoids from the rootstalk of Sophora flavescens Ait., we achieve three monomeric compounds. Establish a HPLC method for determination of trifolirhizin in Sophora flavescens Ait. for the first time. Thismethod is simple, rapid and reliable.This thesis includes three parts as follows:Part â…  : Review. A review of the progress in chemical composition , pharmacological research in Sophora flavescens Ait. and high-speed counter current chromatography were given.Part â…¡: Extraction and separation of the flavonoids from the rootstalk of Sophora flavescens Ait. The medicinal material power was extracted by boiled water, and then the ethanol was added to water solution for depositing some impurities and by circumfluence of organic solvent. The ethyl acetate was added to extract the water solution. The silica gel column chromatography and it with decompression were used to separate the part of ethyl acetate in turn. At last, three monomeric compounds were gained from the two column in all. Two of three monomeric compounds were tirfolirhizin and Kushenol O identified by chemical and spectral method, and trifolirhizin is a main constituent of flavonoids in Sophora flavescens Ait. The result showed that the same flavonoids were gained by circumfluence of organic solvent andby boiled water, and then the ethanol was added to water solution for depositing some impurities. But the previous one is simple.Part HI: Quantitative analysis of the flavonoids from the rootstalk of Sophora flavescens Ait. Establish a HPLC method for determination of trifolirhizin in Sophora flavescens Ait. for the first time. The mobile phase was consisted of MeOH-HaO (1:1) and the determination wavelength was at 310 nm. Then the contents of trifolirhizin in different locations, different sources and patent medicine with Sophora flavescens Ait. were determined by this method. The result showed the content of trifolirhizin in root (0.07%~0.38%) distinct above in aerial part (0.006%~0.034%). The average content of trifolirhizin in Xiaoyin Pian is 0.200 mg/piece and the content is stabilization. This method was suitable for determination of trifolirhizin in Xiaoyin Pian.
