Purpose:To evaluate the clinical therapeutic efficacy and safety of Ying Zhong Xiao Point-application in treating mild to moderate Graves' disease.Material and method:Mild to moderate GD patients from March 2018 to March 2019 in Endocrine Ward of Affiliated Hospital of Liaoning University of Traditional Chinese Medicine were elected and randomly divided into treatment group and control group.The patients in control group took methimazole 15?30 mg/time orally,in treatment group took Ying Zhong Xiao for external point-application in addition.The thyroid function and levels of thyroid autoantibodies,thyroid volume and peak systolic velocity were evaluated at 0 weeks,4 weeks,8 weeks and 12 weeks of treatment.The difference of TCM syndrome score was used to evaluate the effectiveness.Blood samples were collected for blood routine test,liver function test and adverse reactions to assess safety.The data were statistically analyzed by SPSS 24.0 statistical software.Results:A total of 51 cases were included in the study,all of which were mild to moderate Graves patients in Endocrine Ward of Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from March 2018 to March 2019.Three cases?1 in the treatment group and 2 in the control group?were excluded during observation follow-up because of missing visits.The observations during the experiment were as follows:1.Thyroid function and thyroid autoantibody titers: There was no statistical difference in serum FT3?FT4?TSH level between the two groups?P>0.05,P>0.05,P>0.05?before the test.Thyroid function in each group were significantly improved?P<0.01,P<0.01,P<0.01?after treatment.Recovery of serum FT3,FT4,TSH level in the treatment group was significantly better than that in the control group?P<0.01,P<0.01,P<0.01?at the 4th and8 th weeks of the test.Serum FT3?FT4?TSH levels were not statistically different between the two groups?P>0.05,P>0.05,P>0.05?at the 12 th weeks.Both groups had significant time effect on serum FT3,FT4,TSH level?P<0.01,P<0.01,P<0.01?.The results of serum FT3,FT4,TSH level showed that the interaction?P<0.05,P<0.05,P<0.05?between groups and timeat the 8th weeks.The interaction between the invisible group and time?P>0.05?in serum FT3 level at the 12 th weeks.Serum FT4,TSH levels showed interaction between group and time?P<0.05,P<0.05?at the 12 th weeks.There was no statistical difference in serum TRAb antibody level between the two groups?P>0.05?before the test.The serum TRAb in two groups decreased significantly?P<0.01,P<0.01?compared with that before treatment.After treatment,there was no significant difference?P>0.05?in serum TRAb between the two groups.2.Thyroid ultrasound: There was no statistical difference in the volume of thyroid between the two groups?P>0.05?before the test.The volume of thyroid in the treatment group decreased significantly?P<0.01?after 12weeks' treatment.And there was no statistical difference?P>0.05?in the control group after the treatment.The thyroid volume in the treatment group was significantly smaller than that in the control group?P<0.01,P<0.01,P<0.01?at each point in time after the start of the test.The results of thyroid volume in treatment group showed significant time effect difference?P<0.01?.There was no time effect?P>0.05?on the reduction of thyroid volume in the control group.The results of thyroid volume showed that the interaction between groups and time?P<0.05,P<0.05?at the 8th and 12 th weeks of the test.There was no statistical difference in the peak systolic velocity of superior thyroid artery between the two groups?P>0.05?before the test.The peak systolic velocity of superior thyroid artery in the two groups was significantly decreased?P<0.01,P<0.01?compared with that before treatment.After treatment,there was significant difference between the two groups?P<0.01?in the peak velocity of systolic velocity of superior thyroid artery.3.TCM Syndrome Score: There was no statistical difference in TCM Syndrome Score between the two groups before entering?P>0.05?.The scores of TCM syndromes in both groups improved significantly?P<0.01,P<0.01?compared with those before treatment.In treatment group,the improvement of TCM syndrome score was better than that in control group?P<0.01?after treatment.The scores of TCM syndrome based on the effective rate,the treatment group 95.83% was better than the control group 75.83%?P<0.05?.4.The dosage of MMI: MMI starting dose was not statistically different between the two groups?P>0.05?.In the treatment group,the dosage of MMI was lower than that in thecontrol group?P<0.05,P<0.05,P<0.05?at each point in time after the start of the test.Both groups had significant time effect?P<0.01,P<0.01?on MMI dosage.The results of MMI dosage showed that the interaction between groups and time?P<0.05,P<0.05?at the8 th and 12 th weeks of the test.Conclusion:Treatment of mild to moderate Graves disease with Ying Zhong Xiao Point-application showed that 4 weeks and 8 weeks worked better than MMI alone.Ying Zhong Xiao Point-application can reduce the dosage of MMI and reduce the incidence of adverse reactions.
