Objective: Licorice is an important bulk Chinese medicinal material with a wide range of applications.It is often used as a medicinal compound in many traditional Chinese medicines.It is called "Guo Lao" in Chinese medicine.Traditional Chinese medicine believes that " The medicine should be prepared before we take it." The preparation of medicine is to ensure the safety of clinical medication and improve the curative effect of medicine.It is an indispensable link in the application of traditional Chinese medicine.There is a difference in efficacy between unprocessed licorice and processed licorice.Therefore,the reasonable selection of licorice processed products is very important for clinical efficacy.Today,unprocessed licorice and processed licorice are commonly used clinically.But whether the processed licorice is used as honey licorice.It should be based on the preparation methods and clinical use of the prescriptions and herbal medicines in the past.In this project,licorice was processed with clear water,and its main active ingredients were compared.The HPLC fingerprints of licorice before and after processing were studied to determine its detection method.The cytological method was used to compare the cold and heat properties of raw licorice,honey,and water.As the basis,determine the best water-burning process.This subject provides reference and basic research for the follow-up research on other processing technology of licorice and clinical rational selection of licorice varieties,and further development and utilization of corresponding medicinal material resources.Methods:?1?Reviewing the documents about the herbal medicines,medical classics and related literatures,sorting out and analyzing the historical processing methods of licorice,and sorting out the records of licorice processing methods in previous editions of the pharmacopoeia and the regulations on processing technology of licorice decoction pieces processed in all parts of the country.This article summarizes the processing history of licorice,and clarifies the development process of licorice.?2?Select a single factor experiment and take a comprehensive scoring method with the content of glycyrrhizin and glycyrrhizic acid as the evaluation index,and optimize the key process parameters such as the amount of water added to the licorice pieces,moistening time,cooking temperature,and cooking time.Orthogonal experiments were designed to determine the best processing technology of Licorice.?3?Select 18 batches of licorice pieces of different origins or producers,and process them according to the preferred processing technology of licorice root.The HPLC method was used to establish fingerprints of licorice and licorice respectively,and the differences in chemical composition between the two were compared.?4?According to the previous data compilation,the drug properties of licorice have changed after being burned.Therefore,the cell proliferation experiment was used to compare the effects of raw licorice,honey and water on the proliferation of RAW264.7 cells in vitro.Three kinds of licorice cannon were observed under an inverted microscope.The effect of the product on the morphological characteristics of the cells was used to compare the cold and heat properties of the three licorice processed products.Results:?1?According to research,from ancient times to the present,there are many processing methods of licorice.Processing methods without additives include braised,fried,processed,and simmered;methods of manufacturing with accessories include wine,vinegar,salt,oil,Ginger,honey,water,bile and other methods.There are 9 editions of the published pharmacopoeia that document the processing of licorice,and 24 provinces and cities nationwide have recorded the processing of licorice.?2?Through the test,the amount of water added?A?,moistening time?B?cooking temperature?C?and cooking time?D?have no significant effect on the processing of licorice.It can be seen intuitively by comparing the extremely poor R values.,The degree of influence is B> C> D> A.The best process for water-burning licorice is A1B3C3D1,that is,add pure water to the licorice slice,mix well,moisten for 3 h,place in a pot,and fry at 160 ? for 6 min.Cool and sieve to remove debris.Add 20 kg of pure water per 100 kg of licorice.?3?The HPLC method was used to establish the fingerprints of Glycyrrhiza uralensis and Glycyrrhiza uralensis for the Kromasil 100-5-C18?4.6mm × 150 mm,5?m?chromatographic column;the mobile phase was acetonitrile?A?-0.05% phosphoric acid solution?B?,gradient elution?0? 8min,19% A;8?35 min,19%?50% A;35 36 min,50%? 100% A?;detection wavelength 237nm;volume flow 1mL / min;column temperature 30 ?;the injection volume is 10?L,this method is stable and feasible.The comparison fingerprints of licorice raw and water-burned products were compared.The relative retention times of licorice raw and water-burned products were 0.210?peak 1?,0.237?peak 2?,0.459?peak 3?,and 0.537?peak 4?respectively.,0.575?peak 5?,0.604?peak 6?,0.615?peak 7?,0.657?peak 8?,0.720?peak 9?,0.785?peak 10?,0.836?peak 11?,0.853?peak 12?,0.932 There are 17 common peaks around?peak 13?,0.967?peak 14?,1.000?peak 15?,1.071?peak 16?,and 1.094?peak 17?.Three common peaks were confirmed by comparison with the reference,peak 2?6.8 min?was glycyrrhizin,peak 8?19.0 min?was glycyrrhizin,and peak 15?28.9 min?was glycyrrhizin.The similarity of licorice in different batches and places of origin is high.?4?When the drug concentration was 200 ug / mL,the inhibitory rates of the three processed products on the RAW264.7 cell proliferation in vitro were all negative,showing the promotion of cell growth.The inhibitory effects of licorice raw product and honey-flavored product on RAW264.7 basically increased with increasing concentration,and there was a significant concentration-dependent relationship.Glycyrrhiza uralensis?200-5400 ?g / mL?promotes the growth and proliferation of RAW264.7.Conclusion: Through the collection and processing of licorice,it was found that in ancient times,there was a method of drenching licorice with water and roasting it with fire.Based on this topic,orthogonal design L9?34?was used to determine the best processing technology of water-burned licorice,and the HPLC method was used to establish the fingerprint of water-burned licorice.CCK8 method was used to evaluate the cold and heat medicine properties of licorice raw products,water-burned products and honey-burned products.
