Objectives:The objectification and quantification of traditional Chinese medicine(TCM)property is a hot and difficult issue in the study of TCM property theory.It is an urgent problem to be solved in the process of realizing the modernization of TCM property.This project is based on the theory of biophoton emission coherence,using biophoton emission detection technology,though the Scenedesmus as a biological indicator,to explore the biophoton quantization index for characterization processed products and compound prescription of TCM property,thus laying the foundation for the quantification of TCM property.Methods:Detect the biophoton emission of different processed products and several classical compound prescription of TCM using Scenedesmus as a biological indicator,and the experimental data is fitted by the"Gu parameter"model,to seek the biophoton index that can quantitate the differences of different processed products and compound prescription of TCM.HPLC method is used to detect the content of major active ingredients of different processed products of TCM,to explore the correlation between biophoton index and component content.Animal experiments to verify the cold and hot drug properties of processed products of TCM used in the experiment.Contents and results:1.The experimental conditions optimization of Scenedesmus as a biological indicator.Firstly,select the optimal time interval of 0.1s by comparing the fitting degree of the“Gu parameter”of the delayed luminescence(DL)data of the Scenedesmus at different time intervals.Secondly,compare the DL parameters in the different concentration of Scenedesmus.The suitable concentration range of the Scenedesmus is found to be(2.5~3.5)×10~7 cells/mL.In actual experiments,the concentration of the Scenedesmus we used is 3.0×10~7 cells/mL.Finally,the optimal growth stage of Scenedesmus is determined to be 40~60 days by comparing the DL parameters of Scenedesmus at different growth stages.2.Study on property of TCM processed products.(1)Taking the Rehmanniae Radix and Rehmanniae Radix Praeparata as examples,the best dosing amount of the decocted liquid of processed products of TCM is determined to be 100μL.(2)Taking the Arisaematis Rhizoma Preparatuma as examples to verify that the EDL linear fitting slope k is a more stable parameter than the NDL average intensity.(3)Detecte the DL and contents ofβ-sitosterol,bilirubin and bile acid of the Arisaematis Rhizoma Preparatuma and Arisaema Cum Bile(each 6 batches).The results show that EDL linear fitting slope k of the Arisaematis Rhizoma Preparatuma is significantly higher than the Arisaema Cum Bile,i.e.,the slope k of warmed medicine is higher than cold medicine.There is a significant negative correlation between the slope k and the content ofβ-sitostero,and a negative correlation trend with bilirubin and bile acid.(4)Detecte the DL and contents of catalpol,acteoside and 5-HMF of the Rehmanniae Radix and Rehmanniae Radix Praeparata(each 6 batches).We find that EDL linear fitting slope k of the Rehmanniae Radix is significantly lower than the Rehmanniae Radix Praeparata,i.e.,the slope k of warmed medicine is lower than cold medicine,the slope k is only significantly negatively correlated with the content of 5-HMF.The results of mice gavage experiment show that the serum MDA content and liver ATPase activity in mice given Rehmanniae Radix are significantly higher than those in Rehmanniae Radix Praeparata.The results indicate that the quality of Rehmanniae Radix and Rehmanniae Radix Praeparata is poor,and the cold and hot medicinal property may be changed.(5)Detecte the DL and contents of main active ingredient of the Glycyrrhizae Radix Rhizoma,Glycyrrhizae Radix Rhizoma Praeparata Cum Melle,Polygoni Multiflori Radix and Polygoni Multiflori Radix Praeparata(each 6 batches).There is no significant difference in the EDL linear fitting slope k between the raw product and the product,and there is no significant correlation between the slope k and its main active ingredient content.3.Study on property of TCM compound prescription and TCM in its.Firstly,taking Coptidis Rhizoma and Euodiae Fructus as examples,the best dosing amount of the decocted liquid of compound prescription of TCM is determined to be 20μL.After that,the DL is detected of Scenedesmus after adding the decoction liquid of the Sini Decoction,Zuojin Pill,Anti-Zuojin Pill,Mahuang Decoction,Maxinshigan Decoction and TCM within its.The EDL linear fitting slope k of the warming medicine and compound prescription was significantly higher than cool medicine and compound prescription.Conclusions:The EDL linear fitting slope k is a quantifiable biophoton index that can quantify the difference between the different processing produces and compound prescription of TCM.The EDL linear fitting slope k of the warming medicine is significantly higher than cool medicine.This study provides a new idea and method for the modernization of TCM property.
