Objective:To explore the modeling method of Ganqichengpi diarrhea,and investigate the effect of Tongxieyaofang prescription on mice with Ganqichengpi diarrhea caused by senna leaf-binding clip-tail method,and explain the action mechanism of Tongxieyaofang prescription in treating Ganqichengpi diarrhea from intestinal function,microorganisms and enzyme activity,and provide theoretical and experimental basis for better understanding of traditional Chinese medicine.Methods:The mice model of Ganqichengpi diarrhea was prepared by senna leaves extract gavage(1 g/m L)and centrifugal tube binding clip tail method(1 h/day)with 7 days.After diarrhea was induced,Tongxieyaofang prescription(5.67g/kg,0.35m L)was administrated twice a day for 3days.After successful molding and treatment,the changes of intestinal propulsion rate and gastric emptation rate in mice were observed by nutrient semi-solid paste gavage,and the changes of blood biochemistry in mice were detected,as well as the contents of 5-ht,d-xylose and body weight in blood samples.At the same time,intestinal contents and mucosa of each group were extracted under aseptic conditions,which for intestinal microbial culture and intestinal enzyme activity analysis.Results:Through the modeling of senna leaves gavage and centrifugal tube binding clip tail method,model mice have increased loose stool frequency which water-like appearance,and appear irritable and aggressive,weight loss significantly(P<0.01),the content of D-xylose and GLU was reduced significantly(P<0.05),and the bowel advancing rate was increased(P<0.05),the model group mice had no effect on TG and LDL-C(P>0.05);after the treatment of Tongxieyaofang prescription,the Chinese medicine intervention group healed quickly,the mood changes and the degree of diarrhea were improved significantly.The serum of 5-HT,D-xylose content and small bowel advancing rate returned to normal levels(P>0.05),and the GLU(P<0.05)was significantly higher than the normal group,while TC was lower than the normal group significantly(P<0.05),and there was a positive correlation between the change in body weight of the Chinese medicine intervention group and the number of days of gastric administration(R~2=0.7774).After modeling,the number of bacteria in the intestine contents of the model group was higher significantly(P<0.01),the amylase activity in the intestinal contents of the model group and the protease,amylase and invertase activities in the intestinal mucosa weie reduced significantly(P<0.05).After 3 days of Tongxieyaofang prescription treatment,the number of bacteria in the Chinese medicine intervention group was significantly reduced compared to the normal group(P<0.05),and the number of E.coli was apparently lower than the natural recovery group(P<0.05),the intestinal of B/E value returned to normal levels(P>0.05);the enzymatic activity in the anterior segment intestine mucosa态midcourse intestine mucosa and intestinal contents recovered slowly,The activities of cellulase of the anterior intestinal mucosa and invertase and lactase activity in the middle segment of the intestine were significantly lower than those in the normal group(P<0.05),But the lactase,sucrose Enzyme and amylase activities of Chinese medicine intervention group were significantly higher than those in the normal group(P<0.05).Conclusion:A mice model of Ganqichengpi diarrhea can be established by senna leaf gavage and centrifugal tube binding clip tail method.The 5-HT,D-xylose content in mice serum and gastric dynamics change can be used to evaluate the model of Ganqichengpi diarrhea mice.The model of Ganqichengpi diarrhea can increase the number of harmful bacteria such as bacteria,and reduce intestinal contents and mucosa amylase activity significantly;Tongxieyaofang prescription can inhibit the growth of bacteria and E.coli,restore the B/E value to the normal range and balance the intestinal environment.Tongxieyaofang prescription have significant effects on lactase,sucrase,and amylase activity of the mice intestinal mucosa,especially in the tail end of intestinal mucosa.
