| Objective:The effects of moxibustion and acupuncture on colonic tissue metabolism of CD rats were studied by 1H-NMR,and the mechanism and difference of moxibustion and acupuncture were explained from the point of substance metabolism.Methods:Twenty four SD rats were randomly divided into four groups:normal group,model group,moxibustion group and acupuncture group.After the model was established successfully,the rats in moxibustion group and acupuncture group were intervened.The acupoints of "Tianshu point" and "Shangjuxu point" were selected for 15 minutes each time and treated once a day for 7 days.The normal group and the model group were fixed in the same way for 15 minutes without moxibustion or acupuncture intervention.The body weight,fecal blood and fecal characteristics were recorded during the experiment.At the end of the intervention,the colonic tissues of rats were collected and the metabolites of colonic tissues were detected by 1H NMR technology.The NMR spectra were normalized by mestrenova software and imported into simca-p 14.1 statistical analysis was carried out by software,and the differential metabolites and metabolic patterns of colonic tissue were analyzed by principal component analysis,partial least square discriminant analysis,VIP value combined with t-test.Results: Groups of rats PCA scoring diagram,it can be seen that CD rats colon tissue metabolism changed,acupuncture and moxibustion on CD in rats colon tissue metabolism are affected,moxibustion colon tissue recovery better effect after intervention OPLS-DA analysis found that:(1)the moxibustion group vs model group has 5: the marked increase in the concentration of metabolites HX(hypoxanthine)Tau(taurine)Gly(glycine)KG(alpha glutaric acid salt)m-I(inositol)alpha Glc(alpha glucose);The concentrations of 5 metabolites were significantly decreased :Leu(leucine),Val(valine),Ile(isoleucine),Tyr(Tyr),Phe(phenylalanine)(2),and in the acupuncture group vs model group,the concentrations of 4 metabolites weresignificantly increased :HX(hypoxanthine),Gly(glycine),Tau(taurine)and-glc(glutamine)Lac(lactate).There were 4 metabolites with significantly reduced concentrations :Val(valine)Ile(isoleucine)Leu(leucine)3-hba(3-hydroxybutyrate).Conclusion:1.Moxibustion and acupuncture had a good effect on improving the diarrhea and pathological changes of the mental state of hair gloss and water consumption in tnbs-induced CD model rats.2.There are mucosal barrier damage and microbial metabolism abnormality in the colon of CD rats,which complement each other.Due to the influence of intestinal inflammation,infection and complications,the intestine is often in the state of intestinal mucosa damage and high catabolism,and a large amount of energy,amino acid,sugar and fat are consumed.Compared with the normal group,the metabolism of amino acid,energy,sugar and bile acid in CD rats was disordered.3.Moxibustion and acupuncture can regulate the metabolism of amino acid,bile acid,sugar and energy.Moxibustion has more obvious regulation on amino acids.The normal content of branched chain amino acids is conducive to maintaining the integrity of intestinal barrier and protecting the permeability of intestinal mucosa.Therefore,moxibustion regulation is more conducive to promoting the repair of intestinal mucosa and the recovery of intestinal barrier function in CD rats.Acupuncture has more prominent regulation on energy metabolism.Glucose and lactate participate in the tricarboxylic acid cycle,while the whole tricarboxylic acid cycle The important physiological function is to produce ATP.The substance metabolism in the tricarboxylic acid cycle promotes the energy metabolism of CD rats,so acupuncture regulation is more conducive to the normal operation of energy metabolism of CD rats. |
