Objective: To investigate the internal mechanism of Taohong Siwu Tang on early fracture healing from the perspective of molecular biology through the analysis of early bone callus by Micro-CT and the migration of Taohong Siwu Decoction-containing serum to bone marrow mesenchymal stem cells(BMSCs).Methods:1.Randomly select 60 male SD rats and randomly divide them into normal group,sham operation group,fracture model group,Taohong Siwu Tang high,middle and low dose group,10 rats in each group.Fracture model group and Taohong Siwutang high,middle and low dose group completed aseptic osteotomy to establish a right femoral shaft fracture model.The sham operation group only cut the skin and then sutured the wound.The normal group did not do any treatment.After the operation,the amount of drug intervention was calculated according to the "equivalent dose ratio table converted from body surface area between humans and animals".Taohong Siwu Tang high,middle and low three groups of decoctions were administered by gavage;normal group,sham operation group,fracture model group and so on Gavage with normal saline for 14 days.On the 7th,10 th,and 14 th day of intragastric administration,3 rats were randomly selected from each group for fracture callus for direct observation and Micro-CT analysis.2.Randomly select 5 male SD rats and use whole bone marrow adherent method to culture BMSCs in vitro.3.Randomly select 20 male SD rats and randomly divide them into normal group and Taohong Siwu decoction high-medium and low-dose group,and continuous gavage On 7 days,1 hour after the last intragastric administration,blood was collected from the abdominal aorta.After centrifugation,the normal group serum and drug-containing high,medium and low serum group were prepared to intervene to culture BMSCs in vitro.Serum group migration effect on BMSCs.Results:1.On the 7th,10 th,and 14 th days after Taohong Siwu Decoction intervened in the fractured rat model,the bone volume and bone volume fraction of Taohong Siwu Decoction in the high,medium and low dose groups were significantly higher than those in the fracture model group(P<0.05).The intervention effect of the dose group was better.2.Through the cell scratch test,the migration rate of BMSCs in the drug-containing high,medium and low s/erum group was significantly higher than that in the normal serum group(P<0.05);in the migration chamber experiment,the number of cells migrated to BMSCs in the drug-containing high,medium and low serum group was higher than that in the normal group(P<0.05).Conclusion:1.Taohong Siwu Tang can promote early fracture healing in fractured rats.2.The drug-containing serum of Taohong Siwu Tang can promote the migration of BMSCs.3.Taohong Siwu Decoction promotes early fracture healing,in whichthe internal mechanism may be to promote the migration of BMSCs to the fracture ends,thereby promoting the growth of callus and accelerating the healing of fractures.
