Effect Of Buganjian Yaofang On MMP-3 And TIMP-1 In Rat Lumbar Intervertebral Disc Degeneration

Object:Investigating the effect of the MMP-3?TIMP-1 of Bu Gan Jian Yao Fang on the degeneration of lumbar intervertebral disc in rats.Methods:75 SD rats Will be randomly selected 60 as a module,the remaining 15 were only for control.Building group was given in the rat lumbar(L4/5)altered back mould processing,after the success of the building were randomly divided into: model group,Chinese medicine group,Doxycycline,Yao Bi Tong group,15 in each group,were given saline,Bu Gan Jian Yao Fang,doxycycline,Yao Bi Tong group,the control group only exposed intervertebral disc,not to be degenerated,feeded with physiological saline,Respectively in the postoperative before the intervention of drug,20 d,and 40 d day packet collection segment disc tissue(L4/5)in the rats,specimens were stained with HE under a200 x ptical microscope,Reverse transcription-polymerase chain reaction(RT-PCR)method to detect intervertebral disc tissue of MMP-3 m RNA and TIMP-1 m RNA content,immunohistochemistry(IHC)method in the detection of intervertebral disc tissue MMP-3,the content of TIMP-1.Results: 1.The rat lumbar disc tissue after builded in the microscopic observation presents degenerative change before drug intervention,the rat lumbar disc tissue repaired after drug intervention;2.The number of TIMP-1,MMP-3 content expressing are increase in the rat lumbar disc tissue after builded,a significant reduction in the content of expression after drug intervention,TIMP-1MMP-3 / TIMP-1 ratio also fell,the effect is quite between traditional Chinese medicine group and doxycycline group,and is superior to the Yao Bi Tong group.Conclusion: 1.The use of acupuncture in the rat lumbar could be successfully in made lumbar degeneration model,and model the pathological change of state is similar to human,the animal model of the expression of MMP-3 and TIMP-1.2.Bu Gan Jian Yao Fang and doxycycline can lower lumbar disc degeneration in rats model of the expression of MMP-3 and TIMP-1,has therapeutic effect on lumbar degeneration,the effect is quite between Bu Gan Jian Yao Fang anddoxycyclinein the reatment of lumbar intervertebral disc,Bu Gan Jian Yao Fang can reduce the ratio of MMP-3 / TIMP-1,lumbar degenerative diseases and MMP-3 / TIMP-1 also was a significant relationship,the ratio of MMP-3 / TIMP-1 ratio is higher that lumbar degeneration,the lower the ratio,or the closer it gets to the normal lumbar disc degeneration is not serious or recovered well.
