Background:Moxibustion and acupuncture therapy are the two most representative external treatment methods of traditional Chinese medicine.Their clinical curative effect is not only deeply respected by their people,but also recognized and widely used by the international community.The basic principle of the role of moxibustion therapy is mainly to give the local acupoints or meridians of the body warm stimulation by means of the temperature generated when the moxa burning,and the effective stimulation of the body by acupuncture is essentially the needle after the needle into the surrounding the different forms of mechanical force in the organization.When the moxibustion and acupuncture operations are performed,the healer acts on the patient's body with different forms of heat and force to reach the patient's body.The purpose of disease prevention and control is achieved by clearing the meridians,regulating organs and balancing yin and yang.In the previous period,our research group used local acupuncture and moxibustion methods to stimulate local Zusanli acupuncture points with different strengths.The test was performed using an antibody chip that could detect 308 changes in protein expression.The results showed that both acupuncture and moxibustion can promote "Zusanli"acupoint increased local protein synthesis,and moxibustion promoted the expression of stimulated local protein stronger than acupuncture stimulation,which revealed the different effect mechanisms of acupuncture and moxibustion from the molecular level,but different forms of heat and force.What is the basis of the cellular behavior of the changes in protein expression after stimulation,and what are the characteristics of the acupuncture and moxibustion effects of these two different stimuli?These problems are not only the core of the research on the therapeutic effect of acupuncture and massage,but also the key to choose the most appropriate acupuncture and moxibustion method in clinical practice.Objective:To explore the influence of heat and force on the viability of mesenteric fascial fibroblasts,and to explore the non-neurological mechanisms of acupuncture and moxibustion from the perspective of cell biology,and to explore the characteristics and differences of moxibustion and acupuncture.To provide a new experimental basis for a comprehensive understanding of the biothermy and mechanical mechanisms of acupuncture treatment.Methods:Vesicular connective tissue fibroblasts were obtained in vitro by enzymatic digestion.The cells were passaged and amplified and identified.The cells were seeded and divided into groups and administered to different groups of cells at different temperatures(37?,39?,41).(C,43?)and different pressure(Okpa,50 kpa,100 kpa,200 kpa)stimulation,cell viability analyzer was used to measure cell viability,diameter and roundness of the three indicators of cell viability,and all data using SPSS 17.0 Statistical software performs factorial analysis of variance.Result:1.In vitro stimulation with different temperatures can increase the viability of fibroblasts and have a significant effect on cell viability,diameter,and roundness,and they are statistically significant:(1)Effect on cell viability:? There was a significant difference in the viability of fascial fibroblasts with different temperature stimulation intensity(P=0.000<0.001),indicating that the temperature had a statistically significant effect on the cell viability and increased the viability of the cells;? Different times of stimulation had a significant difference in the viability of fascial fibroblasts(P=0.000<0.001),indicating that the number of stimulations was different,and the effect on cell viability was also statistically significant,which increased the viability of the cells;?There was a significant difference in the viability of fibroblasts between the stimulation temperature and the number of stimulation times(P=0.001<0.01),indicating that there was a statistically significant interaction between factor temperature and number of factors,that is there is interaction between temperature and frequency;?Among each group of single simulation,the cell viability in 41 ?group is the highest and the cell viability in 43? group come to the second;? Among each group of multiple stimulation,the cell viability in 41 ? group is the highest and the cell viability in 3 9? group come to the second;?Among each group of single and multiple stimulation,the cell viability in 41? group of multiple stimulation is the highest.(2)Effect on cell diameter:? Different temperature stimuli intensity had significant difference in cell diameter of fascial fibroblasts(P=0.000<0.001),indicating that temperature had a statistically significant effect on cell diameter and increased cell diameter;?Different times of stimulation had a significant difference in the diameter of fascial fibroblasts(P=0.000<0.001),indicating that the number of stimulations was different,and the effect on cell diameter was also statistically significant,resulting in an increase in cell diameter;?There was a significant difference in the diameter of fibroblasts with the superposition of stimulation temperature and stimulation times(P=0.001<0.01),indicating that there was a statistically significant interaction between factor temperature and number of factors,that is there is interaction between temperature and frequency;?Among each group of single simulation,the cell diameter in 41? group is the highest and the cell diameter in 39? group come to the second;? Among each group of multiple stimulation,the cell diameter in 39? group is the highest,the cell diameter in 41?group come to the second and in 43? group is the smallest;? Among each group of single and multiple stimulation,the cell diameter in 41? group of single stimulation is the highest.(3)Effect on cell roundness:?There was a significant difference in the roundness of fascial fibroblasts with different temperature stimulation intensity(P=0.009<0.01),indicating that the temperature had a statistically significant effect on the cell roundness and increased the roundness of the cells;? Different times of stimulation had a significant difference in the roundness of fascial fibroblasts(P=0.018<0.05),indicating that the number of stimulations was different,and the effect on cell roundness was also statistically significant,which increased the roundness of the cells;? There was a significant difference in the roundness of fibroblasts between the stimulation temperature and the number of stimulation times(P=0.001<0.01),indicating that there was a statistically significant interaction between factor temperature and number of factors,that is there is interaction between temperature and frequency;? Among each group of single simulation,the cell roundness in 41? group is the highest and the cell roundness in 43? group come to the second;? Different times stimuli showed a slight increase in the roundness of fibroblasts,but there was no significant difference(P=0.878>0.05),indicating that different times stimuli had no statistical significance on the cell roundness;?stimulation pressure and stimulation times two factor superposition has no significant difference to the roundness of the fascia fibroblasts(P=0.881>0.05),it shows that the interaction between factor pressure and the number of factors is not statistically signifi cant,that is there is no interaction between the pressure and the times;?Among each group of single simulation,the cell roundness in 100kpa group is the highest and the cell roundness in 200kpa group come to the second;?Among each group of multiple stimulation,the cell roundness in 100kpa group is the highest and the cell diameter in 200kpa group come to the second;?Among each group of single and multiple stimulation,the cell roundness in 100kpa group of single stimulation is the highest.Conclusion:(1)Within the scope of stimulation of this study,whether temperature stimulation or pressure stimulation,the activity of fibroblasts can be increased overall.(2)Temperature stimulation stimulates the viability of fibroblast cells more effectively than pressure stimulation.This may explain the principle of acupuncture and moxibustion in cell biophysics from the part of non-neurological mechanisms.That is "the acupuncture is not what the moxibustion is suitable for".
