Purpose:In this experiment,urticaria model rats were used as subjects,the serum total IgE level and skin morphological changes in locus coeruleus region were taken as test indexes to observe the therapeutic effect of acupuncture at "Quchi" and "Xuehai" points on urticaria;To explore the mechanism of acupuncture at "Quchi" and "Xuehai" points on urticaria rats,mast cell degranulation,release of histamine and serotonin,and protein expression differences of P-Lyn/Lyn and P-Syk/Syk in the skin tissue of locus coeruleus region were used as test indicators.To provide theoretical and experimental basis for future clinical research.Material and method:In this experiment,10 male SD rats of 8 weeks old,weighing200-220 g,1 ml of BCG vaccine diluent and 1% egg albumin physiological saline solution were injected into the hind legs,0.5 ml on each side.After 12 days(IgE serum peak),blood was taken from the femoral artery,serum was separated and mixed,and stored at-20 ? until use.Secondly,40 8-week-old male SD rats,weighing 200-220 g,were randomly divided into normal control group,model control group,acupuncture group(Quchi,Xuehai)and positive drug group,with 10 rats in each group.The acupuncture group used acupuncture(0.25mm×13mm)to acupuncture the rat "Quchi" acupoint,the depth of the straight thorn is2-4mm;the "Xuehai"acupoint,the depth of the straight thorn is 2-4mm,the needle is kept for30 minutes,once a day,continuous treatment 14 days.The positive drug control group was given 0.2 mg of loratadine dissolved in 1 ml of 0.9% sodium chloride solution for continuous treatment for 14 days.The normal control group and the model control group were not treated with any intervention.At this time,the previously stored anti-oval serum was used to induce passive skin allergy in rats to establish a model of chronic urticaria.The skin tissue of the abdominal arterial blood and blue spot of the rat was taken.(1)The effect of serum total IgE levels in rats was observed by enzyme-linked immunosorbent assay;(2)The morphology of skin tissue in the blue spot of rats was detected by HE staining;(3)To observe the effect of degranulation of mast cells in skin tissue of rats with blue spot by using toluidine blue staining;(4)Immunohistochemical method was used to observe the release of histamine and serotonin from the skin tissue of rats with blue spot and the expression of P-Lyn/Lyn and P-Syk/Syk proteins.And carry out statistical analysis.Results:1.Results of acupuncture on serum total IgE levels in rats The serum total IgE level in the model control group was significantly higher than that in the normal control group(P<0.05).The total serum IgE content in the acupuncture group and the positive drug group was lower than that in the model control group.There were significant differences between the two groups(P<0.05).There was no significant difference between the acupuncture group and the positive drug group,and there was no statistical significance.2.Results of acupuncture on skin morphological changes in rat blue spot area The normal control group showed that the skin epidermis was thin and normal,the cells were arranged closely and orderly,the structure was clear,and no edema was observed.The model control group showed that the epidermis was thickened,with parakeratosis or hyperkeratosis.The dermis was swollen and deformed,the small invasive vasculature was severely infiltrated,the basal cell nucleus was divided,and the vascular edema was dilated.The positive drug group showed a slight thickening of the epidermis,slight parakeratosis,mild edema of the dermis,occasional inflammatory cells,and mild edema of the blood vessels.The damage was significantly reduced compared with the model control group.The acupuncture group showed that the skin epidermis was slightly thickened,the structure was basically intact,the cell gap was slightly mild edema,the dermal edema was not obvious,and there were occasional inflammatory cells.The damage was significantly reduced compared to the model control group.3.Effect of acupuncture on degranulation of mast cells in skin tissue of rat blue spot Most of the mast cells in the normal control group were intact,and there was no degranulation.The cytoplasm was stained deep.In the model control group,the mast cells were swollen and deformed,the cell membrane was broken,the contour of the cell edge was irregular,and the particles were detached,and the cytoplasm was stained light.There was no significant difference in mast cells between the acupuncture group and the positive drug group,and the degranulation phenomenon of mast cells was less.The color of the cytoplasm was lighter than that of the normal control group,but it was deeper than the model control group.4.Results of acupuncture on the release of histamine and serotonin in skin tissue of rat bluespot Compared with the normal control group,the expression of histamine and serotonin in the other groups were significantly increased.Compared with the model control group,the expression of histamine and serotonin in the acupuncture group and the positive drug group were all weakened,and there were significant differences(P < 0.05);there was no significant difference between the acupuncture group and the positive drug group.5.Effects of acupuncture on the expression of P-Lyn/Lyn and P-Syk/Syk proteins in the skin tissue of rat blue spot Compared with the normal control group,the protein expression of P-Lyn and P-Syk in the other groups was significantly increased;Compared with the model control group,the protein expressions of P-Lyn and P-Syk in the acupuncture group and the positive drug group were all decreased,and there were significant differences(P<0.05).There was no significant difference between the acupuncture group and the positive drug group.Conclusion:1.Acupuncture can inhibit the total serum IgE level in rats and alleviate the pathological changes of skin tissue in the blue spot area of rats,indicating that acupuncture can inhibit type I hypersensitivity reaction and also prove the therapeutic effect of acupuncture on urticaria.2.Acupuncture can reduce the degranulation of mast cells in the skin tissue of the blue spot,thereby inhibiting the release of inflammatory mediators,which may be one of the mechanisms of acupuncture treatment of urticaria.3.Acupuncture can reduce the release of active mediators such as histamine and serotonin in the skin tissue of the blue spot,and reduce the inflammatory response,which may be achieved by inhibiting the degranulation of mast cells.4.Acupuncture can reduce the protein expression levels of P-Lyn and P-Syk in the skin tissue of the blue spot,inhibit the phosphorylation of Lyn and Syk,thereby preventing the signal from being transmitted to the downstream pathway,inhibiting the degranulation of mast cells and inhibiting the release of inflammation.The sexual medium,in turn,inhibits the occurrence of type I hypersensitivity,and achieves the purpose of treating urticaria.This may be an in-depth discussion of the mechanism of acupuncture treatment of urticaria.
