Part One: Inhibition of ocular surface bacteria by Chinese medicine and Fenjian Granules and determination of MICPurpose :The antibacterial effects of nine Chinese medicinal materials(galla chinensis,coptis chinensis,scutellaria baicalensis,schisandra chinensis,honeysuckle,cornus officinalis,bupleurum chinensis,pulsatilla chinensis,eucommia ulmoides)and their decoction free granules on two kinds of eye surface bacteria were studied.And the minimal inhibitory concentration(MIC)of several free-decoction granules against staphylococcus aureus.To explore the inhibitory effects of several decocting solutions of traditional Chinese medicine and their corresponding non-decocting granules on staphylococcus aureus and pseudomonas aeruginosa.And other Fried granules on the MIC of staphylococcus aureus.Material and method:According to the related research on traditional Chinese medicine antibacterial drugs,9 kinds of traditional Chinese medicine decoction and its corresponding anti-fried granules were selected,and two kinds of bacteria(S.aureus,Pseudomonas aeruginosa)isolated from the patient's ocular surface were placed.In the blood plate,the conventional Chinese medicine susceptibility paper agar diffusion method was used to carry out the bacteriostatic test,the length of the inhibition zone was measured,and the bacteriostasis results of each group were compared,and the experiment was carried out 3 times,and the sputum was taken for statistical analysis.According to the antibacterial results of several traditional Chinese medicines and Fenjian granules,the next experiment was carried out.The double-tube dilution method was used to treat Chinese medicinal herbs,such as gallnut,berberine,astragalus,schisandra,honeysuckle,hawthorn,Bupleurum and Pulsatilla,to Staphylococcus aureus.Determination of bacteriostatic MIC.Results:The effects of berberine,gallnut and astragalus on Staphylococcus aureus were significantly better than those of Schisandra,Honeysuckle,Hawthorn,Bupleurum,Pulsatilla,Eucommia and Staphylococcus aureus(p<0.05);Galla Chinensis,Astragalus and Rhizoma Coptidis against P.aeruginosa The bacteria act as a mediator;Schisandra,Hawthorn,Bupleurum and Eucommia have no obvious antibacterial activity against Pseudomonas aeruginosa.For the decoction group and the Chinese medicine Fenjian granule group of the same drug,there was no significant difference between the groups of each drug(p>0.05);for the inhibition of Pseudomonas aeruginosa and Staphylococcus aureus,between each group,The effect on inhibition of S.aureus was significantly better than that against Pseudomonas aeruginosa(p<0.05).The MIC of Galla Chinensis for Staphylococcus aureus is 0.975~1.95mg/m L;the MIC of Astragalus membranaceus is 1.95~3.9 mg/m L;the MIC of Coptis is 1.95~3.9 mg/m L;the MIC of Schisandra is 7.81~15.63 mg/m L;Pulsatilla The MIC was 3.9~7.81 mg/m L;the MIC of honeysuckle was 15.63~31.25 mg/m L;the MIC of Bupleurum was 62.5~125 mg/m L;and the MIC of Hawthorn was 31.25~62.5 mg/m L.Conclusion:80% strain of levofloxacin combined with baicalin showed synergistic effect,70% of ceftazidime combined with baicalin showed synergistic effect;chlorogenic acid combined with levofloxacin,ceftazidime 70% strain showed additive effect,which may be used for future clinical treatment of ocular surface Staphylococcus aureus To synergy.Part Two: The second part of Chinese medicine monomer combined with western medicine antibiotics on the inhibition of ocular surface Staphylococcus aureusPurpose :Bacterial sacs and meibomian glands in the ocular surface structure can be found in bacteria,and Staphylococcus aureus is a bacterium that can be hidden in the ocular surface.Once it enters the eye,the consequences are serious.Conjunctival sac is the necessary way for internal eye surgery.During surgery,the eyelids are squeezed and stimulated,so that the meibomian gland secretions are discharged into the conjunctival sac.If bacteria are present in the meibomian glands,this will inevitably affect the conjunctiva.The sac causes pollution and greatly increases the chance of infectious endophthalmitis.With the widespread use and even abuse of antibiotics,bacterial resistance has also emerged.Screening of bacteriostatic active ingredients from traditional Chinese medicine monomers has sought a new way to inhibit Staphylococcus aureus.This study was to investigate the antibacterial effect of baicalin,chlorogenic acid combined with levofloxacin and ceftazidime on the inhibition of S.aureus.Material and method:Through the conclusion of experiment,the chlorobacterin extract and the astragalus extract baicalin were selected for antibacterial test.The combined antibacterial effect of baicalin,chlorogenic acid and levofloxacin and ceftazidime was determined by checkerboard microdilution method.The MIC was recorded.Results: After the combination of levofloxacin and chlorogenic acid,the MIC of chlorogenic acid to experimental bacteria decreased from 2~4 mg/m L to 500~1000?g/m L;the MIC of levofloxacin to experimental bacteria decreased from 4~8?g/m L to 2-4?g /m L.(80%strain additive effect)After the combination of levofloxacin and baicalin,the MIC of baicalin decreased from256~512?g to 32~64?g/m L;the MIC of levofloxacin to experimental bacteria decreased from4~8?g/m L to 1~2?g/m L.(80% strain synergistically)MIC of ceftazidime combined with baicalin: After ceftazidime combined with baicalin,the MIC of baicalin to experimental bacteria decreased from 256~512?g/m L to 64~128?g/m L;the MIC of ceftazidime to experimental bacteria decreased from 32~64?g/m L To16~32?g/m L.(70% strain synergy)MIC of ceftazidime combined with chlorogenic acid inhibition: After the combination of levofloxacin and chlorogenic acid,the MIC of chlorogenic acid to experimental bacteria decreased from 2~4 mg/m L to 500~1000?g/m L;the MIC of ceftazidime against experimental bacteria was 32~64?g/m L was reduced to 16~32?g/m L.(70% strain additive effect)Conclusion:The combination of baicalin and chlorogenic acid with levofloxacin and ceftazidime can reduce the respective MIC.
