Purpose:Establish and perfect the bacterial biofilm model for resistant Staphylococcusaureus.Investigation of the inhibition effect of Radix et Rhizoma Rhei on resistance bacteriabiofilm. Between the investigated compounds in combination on the role of bacterial biofilms.Method: In this study, micro-culture96-well plates using Crystal violet, staining,microplate determination of OD values. To establish bacteria biofilm model for the resistancebacteria staphylococcus aureus.SEM and CLSM observation of drug-resistant biofilmmorphology, and further define the film formation of the state. Resistant Staphylococcusaureus biofilm model for drug screening models, co-cultured samples of bacteria and biofilm,investigated through the calculation of inhibition rate and fractional inhibitory concentrationof each sample alone or the combination of bacteria and bacterial biofilms inhibition.Resμlt: Identified the resistance S.Aureus in the formation of biofilms,37℃for24hintegrated microplate reader measured OD values were observed results of SEM and CLSM.Rhein the normal S.Aureus the inhibitory rate of78.17%inhibition rate of resistanceS.Aureus up to62.62%. Screening compounds daphnetin, pyrogallic acid,berberine bisulfateon S.Aureus and bacterial biofilm has a very strong inhibition, the inhibitory rate of coke didnot gallic acid resistance S.Aureus up to71.78%, berberine bisulfate up to72.14%, inhibitionof resistant S.Aureus daphnetin inhibitory rate up to73.22%The combination results:daphnetin, pyrogallic acid, berberine bisulfate and rhein combined to produce a certainsynergistic inhibitory effect on normal resistance S.Aureus, and bacterial biofilm.Conclμsion: With application of SEM and CLSM experiments, the establishment ofdrug-resistant bacterial biofilm model. The role of traditional Chinese medicine compound onbacterial biofilm, to further verify the trace as the feasibility of screening the treatment S.Aureus infectious diseases drug model. Filter out bacteria and bacterial biofilm has a stronginhibitory effect of several anthraquinone in Rheum officinale and with daphnetin, pyrogallicacid,berberine bisulfate applications normal resistance S.Aureus, and bacterial biofilm havesome synergistic inhibition role for the further development of new drug treatment S.Aureusinfectious diseases, scientific basis, to lay a good foundation for further research.
