| Purpose:To explore the effects of replenishing Qi nourishing Yin?detoxication and removing turbidity on NF-?B and TGF-? in diabetic macrovessels rats,so as to understand the mechanism of protecting diabetic macrovessels by the compound prescription of replenishing Qi nourishing Yin?detoxication and removing turbidity.Materials and Methods: 75 healthy male Wistar rats of clean grade were randomly selected as normal control group and fed with common diet according to their body weight;10 rats were randomly selected as AS group and fed with high-fat diet;13 rats were randomly selected as prevention group of traditional Chinese medicine and fed with high-sugar and high-fat diet;the remaining 42 rats were fed with high-sugar and high-fat diet.One month later,a small dose of streptozotocin was injected intraperitoneally to replicate the pathological model of 55 diabetic rats.43 diabetic rats were successfully established.According to the blood glucose level,they were randomly divided into six groups: normal group,model group,positive drug group,prevention group,treatment group and AS group,there are 10 rats in every groups.Drug administration and feeding methods: After the successful establishment of the model,the Chinese medicine treatment group was given high-sugar and high-fat diet and Chinese medicine gavage;the positive medicine group was given high-sugar and high-fat diet and Fosinopril sodium tablet gavage;the model group was given high-sugar and high-fat diet and normal saline at the same dose;the normal group was given normal diet and normal saline at the same dose;the prevention group was given high-sugar and high-fat diet and Chinese medicine gavage;the AS group was given high-fat diet and the same dose of saline.Give the rats drug once a day.Drinking water freely during the experiment and collecting specimens after 10 weeks.The general state of rats before and after treatment,pathological changes in the arterial wall?the changes of blood glucose level and lipid before and after the experiment were observed,and the contents of NF-?BmRNA and TGF-?mRNA and the expression of NF-?B protein and TGF-? protein.Result: After modeling,except AS group,the blood glucose level of rats in each group was significantly different from that of normal control group(p < 0.01).Afterr treatment,the blood glucose of treatment group was lower than that of model group(p < 0.05);the blood glucose of prevention group was lower than that of model group(p < 0.01),but still higher than that of normal group.From Table 1 and Figure 1,it can be seen that the blood glucose of prevention group of Chinese medicine decreased most obviously;the effect of positive medicine on reducing blood glucose was not obvious(p > 0.05),and the difference was not significant(p >0.05).Comparing the AS group and normal group,the blood glucose level of AS group was higher(p < 0.01).The values of TC and LDL-C in TCM prevention group and TCM treatment group decreased significantly(p < 0.01),especially in TCM prevention group.Compared with model group,TCM prevention group could reduce the contents of NF-?BmRNA and TGF-?mRNA,and inhibit the expression of NF-?B protein and TGF-?protein.Conclusion:1.Replenishing Qi nourishing Yin?detoxication and removing turbidity Chinese herbal compound may improve the general state of diabetic macroangiopathy rats,and reduce the blood glucose and lipid levels of diabetic rats.2.Replenishing Qi nourishing Yin?detoxication and removing turbidity Chinese herbal compound may reduce the contents of NF-?BmRNA and TGF-?mRNA and inhibit the expression of nuclear transcription factor NF-?B protein and transforming growth factor TGF-? protein.3.Replenishing Qi nourishing Yin?detoxication and removing turbidity Chinese herbal compound may reduce the damage of diabetic rat's great vessels,and have certain effects on the preventing and treat diabetic macrovascular disease. |
