Purpose:To study the effect of Chinese herbal compound “Yi Tang Kang”on the regulation of GLP-1 and energy metabolism in type 2 diabetic rats with spleen deficiency.For the development and utilization of traditional Chinese medicine compound “Yi Tang Kang”,expand the territory of diagnosis and treatment of “Yi Tang Kang”,and provide a theoretical basis for better service in clinical treament.Materials and Methods: Using 70 healthy male rats,feeding for one week,then feeding with high-sugar and high-fat diet for 4 weeks.A model of type 2 diabetes was replicated by intraperitoneal injection of streptozotocin(STZ),and STZ(45 mg/kg)was intraperitoneally injected in small amount and hingh frequency.After 72 hours of injection,detecting the level of blood glucose of the rats,when it ?16.7mmol/L it was determined to be a successful model.The rats were randomly divided into model group,feeding hydrochloride group(positive drug group),Yi Tang Kang high,medium and low dose group,Yi Tang Kang medium dose combined with metformin hydrochloride group(combined group),10 rats in each group.Yi Tang Kang high,medium and low dose groups were fed the same amount(1ml/100g)of different concentrations of Yi Tang Kang Decoction.Combinating drug group was administered at medium dose of Yi Tang Kang at 9:00 am.Metformin hydrochloride(0.5 mg/kg)was administered at 4 pm,the model group was given the appropriate amount of placebo,and fed with high-sugar and high-fat diet.The other 10 rats as blank group,fed normal food,without any interpretion.The rats diet:water intake and mental state were observed every day,and the blood glucose,body weight and urine volume of the rats were measured every week.After 4 weeks,the rats were killed and the blood was taken to make serum.And the sample was used to measure insulin,glycosylated hemoglobin,glucagon-like peptide-1,superoxide dismutase(SOD),GSH-Px,malondialdehyde(MDA),serum cholesterol(TC),Ester(TG),high density lipoprotein(HDL),low density lipoprotein(LDL)and glycerol in each group.(2)Rat apical tissue and liver tissue were used to measure rat AMPK,Na-K-ATPase,pyruvate,ATP,mitochondrial respiratory chain complex I,mitochondrial respiratory chain complex II,mitochondrial respiratory chain complex Compound III,mitochondrial respiratory chain complex IV,mitochondrialrespiratory chain complex V,and conducting analysis.Result:1.The experimental study found that the body weight of the model group were gradually decreased.The weight loss trend of each treatment group decrease gradually,and after the 4week's treatment,the metformin hydrochloride group and the middle and high dose groups were significantly better than the model group.2.Basal metabolic rate: After 4 week's treatment,the basal metabolic rate of the model group was higher than that of the normal group,and the metformin hydrochloride group and the Yi Tang Kang group had reversal effects,showing a certain therapeutic effect.3.Diabetes-related indicators: blood glucose levels and glycated hemoglobin levels in the model group were higher than those in normal group;blood glucose levels in the metformin hydrochloride group,Yi Tang Kang high,medium and low amount groups began to decrease from the second week after treatment,in the At fourth week,the blood glucose levels of the high,middle and low dose groups of Yi Tang Kang had decreased significantly and were close to the blood glucose levels of normal rats,and glycated hemoglobin was significantly decreased than the model group.The GLP-1 level in the diabetic model rats was significantly decreased than in the normal group.The GLP-1 levels in the metformin hydrochloride group,the Yi Tang Kang high,the middle and low amount groups,and the combination group had significantly reversal effects(p<0.01 or p<0.05).The AC of the model group was significantly increased than that of the blank group(p<0.01),and the other treatment groups were significantly decreased than the model group(p<0.01 or p<0.05).The PKA of the model group was significantly increased than that of the blank group(p<0.01).The PKA of the metformin hydrochloride group,the combination group and the Yi Tang Kang high,middle and low amount groups were significantly decreased than the model group(p<0.05).Insulin in the model group was significantly increased than the normal group(p < 0.01),and no significant effect was observed in each treatment group.4.Energy metabolism in heart: ATP level,LDH enzyme,CPK level,SOD activity,GSH-PX level,complex I,complex III,complex IV levels in the model group were significantly increased than the blank group(p < 0.05);metformin hydrochloride group,beneficial Yi TangKang group and middle amount groups had significant reversal effects on each index(p<0.01 or p<0.05).Energy metabolism in liver: Na+-K+-ATPase and AMPK levels in the model group were significantly dncreased than those in the blank group(p<0.01);each group had a callback effect on Na+-K+-ATPase,and the positive drug group,high and medium dose groups had a callback effect to AMPK.5.Substance metabolism in liver: hepatic glycogen in the model group was significantly increased than the blank group(p<0.01);A-CoA and pyruvate in the model group were significantly increased than those in the blank group(p<0.01);metformin hydrochloride and high-dose groups in Yi Tang Kang group There was a significant reversal effect;combined with the drug groups,Yi Tang Kang low-and medium-amount groups had a callback effect on A-CoA(p<0.01 or p<0.05).6.Substance metabolism in blood: TG and LDLC in the model group were significantly increased(p < 0.01),while HDLC was significantly decreased(P < 0.01).After 4 week's treatment,TG and LDLC were decreased in the metformin hydrochloride group(p < 0.01).At the same time,HDLC were increased(p<0.01);TC in the combination group was decreased than that in the model group(p<0.05);TG and LDLC levels in the high,medium and low dose groups of Yi Tang Kang were lower than those in the model group(p<0.01 or p< 0.05);Compared with the model group,the high-amount and middle-amount groups of Yi Tang Kang had elevated HDLC levels(p<0.01).MDA and ATGL in the model group were significantly increased than those in the normal group(p<0.05 or p<0.01).Compared with the model group,MDA and ATGL were decreased in the positive and high dose groups(p<0.05 or p<0.01).7.Endocrine system: T3,T4,and 17-OHCS in the model group were significantly increased than those in blank group(p<0.01);the metformin hydrochloride group,the combination group,the Yi Tang Kang,and the middle dose group had good callback effects on T4 and17-OHCS.8.Cyclic nucleotide system: cGMP and cAMP in the model group were significantly increased than those in the blank group(p<0.01);metformin hydrochloride and Yi Tang Kang high-dose group were significantly decreased than the model group(p<0.01);cAMP/cGMP in each group had no significant difference.The substance metabolism and energy metabolism oftype 2 diabetes had been enhanced.According to the results of cluster analysis and PCA analysis,it can be seen that the traditional Chinese medicine compound “Yitangkang” can improve the material metabolism and energy metabolism of T2 DM and thus play a good therapeutic role.Conclusion:1.Studies had shown that Chinese herbal compound Yi Tang Kang regulated the disorder of glucose metabolism caused by T2 DM,amend the disorder body's fat metabolism,improved serum GLP-1 level,and played a good therapeutic role in T2 DM.2.The substance metabolism and energy metabolism were improved in T2 DM,and Yitangkang had a significantly regulatory effect.It may be action mechanism of of Yi Tang Kang by regulating the level of substance and energy metabolism in diabetic patients.
