The Effect Of Xiaochaihu Decoction Combined With Exemestane On The Expression Of MMP-2 And E-cadherin In Nude Mice With Breast Cancer

Purpose : In this study,the MDA-MB-435 tumor bearing nude mouse model was established.Researching Chinese Herbal Compound formula-Xiaochaihu decoction and Exemestane on tumor weight.Researching Chinese Herbal Compound formula-Ruyanning and Exemestane on expressions of MMP-2 and E-cadherin of breast tumor-bearing nude mice,to reserch the possiblemechanisms of Ruyanning interfering endocrine therapy.Material and method:(1)preparation of MDA-MB-435 cell suspension: HER-2/nue breast cancer MDA-MB-435 cell line with high growth rate and trypsin digestion was used to form cell suspension.(2)MDA-MB-435 cells in nude mice model: female ovariectomized athymic MDA-MB-435 nude mice inoculated with each right axillary fat pad suspension cells under 0.2m L,inoculation,the formation of human breast cancer MDA-MB-453 xenografts in nude mice were inoculated with the high expression of HER-2/neu,4,the tumor grew to 1cm3,based in aseptic operation under the condition,and then the tumor blocks were inoculated into 1mm3 size,the right armpit fat pad excision in athymic nude mice ovary under sterile conditions,the construction of the nude mouse model,a total of 40 athymic female ovariectomized nude mice were inoculated in 4-5 days after planting,the visible tumor growth can reach 100%nodules,tumor formation,feeding 15 days after modeling success.(3)group and drug delivery;after successful construction of nude mice model of breast cancer,40 nude mice were divided into 4 groups,with 10 rats in each group.The control group: the control group was given 0.9% normal saline 0.4ml/ only /d,once daily gavage.In the exemestane group: 0.3mg /d,daily gavage once;Small Xiaochaihu Decoction and reducing prescription group: "small Bupleurum soup and reducing prescription" 0.4ml/ only /d gavage,daily gavage;Xiaochaihu Decoction plus exemestane group: exemestane group: 0.3mg /d+ "Xiaochaihu Decoction" 0.4ml/ /d by intragastric gavage once daily.The four groups were administered continuously for 21 days.(4)The rats were sacrificed: each group of 10 nude mice was killed at the end of 24 hours after the last Administration(Sixteenth days after the establishment of the model).After the death of the neck,the tumor tissues were stripped,the weight of the tumor was measured and the tumor inhibition rate was calculated,and the tumor tissue was spare.(4)Using Western blot method for the determination of matrix metalloproteinase-2(MMP-2)and E-cadherin(E-cadherin)relative content.The process is shown in figure:Results:1.Tumor weight(g)and tumor suppressor rate blank control group: 1.032±0.870;Exemestane group:0.364±0.165;65.59%Xiaochaihu decoction group:0.566±0.172;47.16%Xiaochaihu decoction +Exemestane group:0.270±0.103;74.74%2.The expression of MMP-2 protein and Ecadherin:(1)The expression of MMP-2:6529.232±128.926,4232.837±72.004,5245.171±100.890,2815.578±267.589?(2)The expression of Ecadherin: 2288.170±150.101,4929.361±187.661,3131.750±169.352,6371.439±125.152?Conclusion:1.The effect of.Xiaochaihu Decoction Combined with exemestane on MDA-MB-435 cells in breast cancer was inhibited.There are synergy.2.Xiaochaihu Decoction Combined with exemestane can regulate the expression of MMP-2and E-cadherin protein level.
