Electroacupuncture On The Expression And Related Mechanism Of NGF And P75NTR In Rats With Cerebral Ischemia-reperfusion Injury

Objective:To observe the effects of electroacupuncture on specific acupoints and the expression levels of nerve growth factor(NGF)and low-affinity neurotrophin receptor p75(p75NTR)in brain tissue of rats with middle cerebral artery embolism and reperfusion,and to investigate whether electroacupuncture points are specific.It has a positive effect on the regeneration of axons and provides a new theoretical and experimental basis for the treatment of ischemic cerebrovascular diseases.Methods:Thirty Sprague-Dawley rats(healthy males)were randomly divided into two groups: sham-operated(6)and model-replicating(24).With reference to Longa's modified suture method,rats in the model-replicating group were subjected to middle cerebral artery embolization and reperfusion,and cerebral ischemia-reperfusion injury was induced in the model-replicating group.According to Longa's model evaluation criteria,neurological function was evaluated in experimental rats.Thirteen successful rats were modeled,6 died and 5 failed.After the model replication,the model successfully replicated rats(13 rats)were randomly divided into 7 rats in the ischemic group(one mouse died during the experiment)and the electroacupuncture treatment group(6 rats).The rats in the electroacupuncture treatment group were given needles.Stabbing Baihui,Yintang and bilateral Zusanli points were stimulated by electroacupuncture.During the treatment,rats in the sham-operated group and the ischemic group did not undergo any intervention.Continuous electroacupuncture intervention for 2 weeks,after the last 24 hours of electroacupuncture intervention,three groups of experimental rats were scored once again for neurological function.Morris water maze test was performed after completion of scoring,and all rats were sacrificed after pulpal dissection.The rat ischemic penumbra was detected.Western blot was used to detect the expression of NGF and p75 NTR protein in the ischemic penumbra.Result:1.Comparing with the sham operation,the neurological function score of the model replication group rats was statistically significant(P<0.01).After the end of treatment,compared with the sham group,the neurological function scores of the ischemic group andthe electroacupuncture group were statistically significant(P<0.01);compared with the ischemic group,the neurological function of the electroacupuncture group rats The score was statistically significant(P<0.01).2.Compared with sham operation,the escape latency of the experimental rats in the ischemic group and the electroacupuncture treatment group was prolonged,the residence time in the target quadrant was shortened after the number of plateaus decreased(P<0.05,P<0.01);In comparison,the evasion latency of the electroacupuncture treatment group rats was shortened,and the electroacupuncture treatment group rats significantly increased after the platform times,and the residence time in the target quadrant was significantly longer(P<0.01).3.Compared with the sham-operated group,the expression of NGF and p75 NTR protein in the brain tissue of the rats in the ischemia group and the electroacupuncture group was significantly higher(P<0.01);compared with the ischemic group,the electroacupuncture group The expression of NGF protein in rat brain was significantly increased,and the expression level of p75 NTR protein was also significantly increased(P<0.01).Conclusion:1.Electroacupuncture Baihui,Yintang,and Zusanli points can significantly improve the neurological deficit scores of rats with cerebral ischemia-reperfusion injury,and have certain protection and positive effect on the ischemic brain tissue.2.Electroacupuncture at Baihui,Yintang and Zusanli can effectively improve the learning and memory ability of experimental rats with cerebral ischemia-reperfusion injury.3.Electroacupuncture at Baihui,Yintang and Zusanli acupoints can promote the expression of NGF and p75 NTR protein in experimental rats with cerebral ischemia reperfusion injury,which may reduce cerebral ischemic injury and promote the regeneration of axons.
