Purpose:By observing the modification of Aconite Decoction on the expression of IL-1?,TNF-? and MMP-13 in serum and articular cartilage of osteoarthritis mouse model.To explore the possible pathogenesis of osteoarthritis and the preventive and therapeutic effects of the modification of Aconite Decoction on osteoarthritis,and provide theoretical basis for clinical treatment and drug research and development.Material and method:60 healthy SPF female Kunming mice were selected as study subjects.According to the random number method,they were divided into blank group,model group,control group,the modification of Aconite Decoction low-,medium-,and high-dose group,10 in each group.Except for the blank group,all other groups of mice were treated with medium meniscectomy(DMM)to establish an osteoarthritis model.One week after the modeling,mice in each group were given intragastric administration.The control group was given wolfberry suspension,the low-,medium-,and high-dose groups of the modification of Aconite Decoction were given low-,medium-,and high-concentration liquids respectively.The normal group and model group were given normal saline for 8 weeks.After 8 weeks,blood was collected from the eyeballs of each group,and the supernatant was centrifuged.After the blood was taken,the mice were sacrificed by the cervical dislocation method,and the knee joints and cartilage tissues were taken.HE staining was used to observe the morphology of articular cartilage in mice in each group.Detection of MMP-13 expression in articular cartilage using real-time quantitative PCR.Results: 1.The results of HE staining of each group of mice showed that the cartilage surface of the blank group was smooth,the cartilage matrix was pale pink,the nucleus was blue,and the cartilage cells were ordered and distributed evenly from top to bottom.The cartilage surface of the model group was not smooth,the number of cells was small,the arrangement was disordered,the stratification of the structure was not obvious,and cell aggregation was observed.The cartilage surface of the control group was relatively smooth,the number of cells increased significantly,and cartilage cells were arranged in an orderly manner.The cartilage surface of the low-dose group of the modification of Aconite Decoction was basically smooth,the arrangement of chondrocytes was neat than that of the model group,the structure of each layer was slightly disordered,and the cartilage cells could be seen on the surface.The structure of each layer of middle dose group of the modification of Aconite Decoction is basically neat.The number of cells in high dose group of the modification of Aconite Decoction increases.2.The detection results of IL-1? and TNF-? in serum of mice showed that the expression of IL-1? and TNF-? in the serum of the model group increased compared with the blank group(P<0.01).Compared with the model group,the expression of IL-1? and TNF-? in the serum of the low-,medium-,and high-dose groups of the control group and the modification of Aconite Decoction decreased(P<0.01).Compared with low dose,medium dose,and high dose of the modification of Aconite Decoction,the expression of IL-1? and TNF-? in serum of mice with medium to low doses was lower than that in low dose group(P<0.01).The expression of IL-1? and TNF-? in serum of mice with high-dose to medium-dose group was decreased(P<0.01).Compared with the control group,the serum levels of IL-1? and TNF-? in the high-dose group of the modification of Aconite Decoction increased(P<0.01).3.The results of MMP-13 expression in mouse articular cartilage showed that the expression of MMP-13 in the articular cartilage of the model group increased compared with the blank group(P<0.01).Compared with the model group,the expression of MMP-13 in the articular cartilage of mice in the low,middle,and high dose groups of the control group and the modification of Aconite Decoction decreased(P<0.01).Compared with low-,medium-,and high-dose groups of the modification of Aconite Decoction,the expression of MMP-13 in the articular cartilage of the mice with medium-dose and low-dose groups was decreased(P<0.01).The expression of MMP-13 in the articular cartilage of mice with high-dose to medium-dose group was decreased(P<0.01).Compared with the control group,the expression of MMP-13 in the articular cartilage of mice with high dose of the modification of Aconite Decoction was increased(P<0.01).Conclusion: 1.The mouse model of knee osteoarthritis was successfully performed using the DMM method.Pathological changes of OA were observed in the cartilage tissue of the knee of the mouse model.The expression of IL-1? and TNF-? in the serum and the expression of MMP-13 in the articular cartilage were increased.Explain that the three are involved in the pathological process of OA.2.The modification of Aconite Decoction with the effect of warming kidney,dispersing stagnation,promoting blood circulation,dispelling dampness and relieving pain can reduce the expression of IL-1? and TNF-? in serum of OA mice and the expression of MMP-13 in articular cartilage and improve the pathological morphology of cartilage.The evidence-empowering machine shows that the traditional Chinese medicine pathogenesis of "deficiency of Han deficiency,coldness and blood stasis" is closely related to the cartilage degeneration mechanism of OA.3.The modification of Aconite Decoction can reduce the expression of MMP-13 in the serum of OA mice model and reduce the expression of MMP-13 in articular cartilage,inhibit the inflammatory reaction,and reduce the damage of various cytokines on chondrocytes and cell matrix to promote cartilage the role of repair.
