Objective: To investigate the effect of Danggui Buxue decoction on hepatic adiponectin and its receptor expression in rats with NAFLD.Methods: The 60 Wistar male rats aged 5~6 weeks were random Ly divided into 6 groups,including control group,model group,rosiglitazone group,Danggui Buxue decoction low-dose group,Danggui Buxue decoction medium-dose group and Danggui Buxue decoction high-dose group.Each group of 10 rats was reared for 14 weeks.Except control group were given by normal diet,All other groups were fed on high-fat and suger diet.The rats in the control group and the model group were given normal saline(6 m L/(kg·d)),rosiglitazone group was given rosiglitazone suspension(4 mg/(kg·d)),and Danggui Buxue decoction(3 g/kg,6 g/kg,12 g/kg)for low,medium and high dose groups.Their body weight was measured every week.Samples were collected after the 14 th week to calculate the liver index,and biochemical indexes such as ALT,AST,TG,TC,LDL-C,HDL-C and FPG in the blood were detected.Paraffin sections of some liver tissues were prepared and stained with HE.The histological changes of each group were observed under the microscope,and the expressions of adiponectin,adiponectin receptor 2 proteins in the liver were detected by Western blot.Results:(1)Compared with the control group,the liver index(liver mass/body mass ratio)increased in the model group(P<0.01).Compared with model group,The liver indexes in the Danggui Buxue decoction medium and high dose groups decreased(P<0.05,P<0.01).(2)Macroscopic observation of the model group showed that hepatic steatosis was severe.Compared with the model group,the hepatic steatosis was alleviated in Danggui Buxue decoction each group,which was less effective than rosiglitazone.(3)The pathological changes of rat liver tissue was observed under optical microscope,the lobular structures was injured in model group,disorganized hepatocyte and hepatic sinus narrow gap compared with the control group,liver cell edema,part of the nucleus,fatty degeneration,lipid drops existed in the form of big cavity,at the same time with inflammatory cells infiltration,necrosis of more;the degree of hepatic lesions in Danggui buxue decoction all groups were improved,showing slightly disordered hepatic lobule structure,irregular arrangement,insignificant cell swelling,a small number of fat vacuoles and no obvious inflammatory infiltration,among which rosiglidone group and Danggui buxue decoction high-dose group had the best effect.(4)Compared with the control group,the blood glucose level in the model group increased significantly(P<0.01),and the Danggui Buxue decoction high-dose group had a certain hypoglycemic effect compared with the model group(P<0.01).(5)the body mass of rats increased steadily,with the largest increase in the model group,and the increase in the drug administration group was smaller than that in the model group,but larger than that in the control group.(6)Compared with the model group,The ALT content of Danggui Buxue decoction low-dose group was decreased(P<0.05);The ALT,AST content in the medium-dose group was decreased(P<0.05),while the difference was significant in the high-dose group(P<0.01).(7)Compared with the control group,the levels of TG,TC,LDL-C were significantly increased(P<0.01),while HDL-C levels were decreased(P<0.01);The TC levels in Danggui Buxue decoction low-dose group was decreased(P<0.05),TG and LDL-C levels were significantly lower,HDL-C levels increased(P<0.01);The levels of TG,LDL-C in Danggui Buxue decoction medium dose group were significantly reduced(P<0.01),TC levels was lower,HDL-C levels increased(P<0.05);The levels of TG,TC,LDL-C in Danggui Buxue decoction high dose group were reduced(P<0.01),HDL-C levels increased(P<0.01).(8)The protein expression levels of ADPN and ADPN R2 in each group were detected by Western blot,which were decreased.Danggui Buxue decoction low-dose group increased adiponectin levels(P<0.05),and at the same time increased the expression of adiponectin receptor 2(P<0.01),and the expressions of adiponectin and adiponectin receptor 2 were increased in the medium-dose and high-dose groups of Danggui Buxue decoction(P<0.01).Conclusion: Danggui Buxue decoction has certain protective effect on the liver of Nonalcoholic fatty liver disease,which may be related to the regulation of glucose and lipid metabolism and the influence of cytokines,etc.The mechanism may be related to the up-regulation of adiponectin and adiponectin receptor 2 protein expressions.
