Purpose:This experimental study with 1-methyl-4-phenyl-5-four hydrogen pyridine(MPTP)induced Parkinson's disease model mice(C57BL / 6)as the research object,observe the different electroacupuncture(EA)treatment for PD model of mice was studied the substantia nigra dopamine neurons phosphorylation c-jun amino terminal kinase(p-JNK),the phosphorylated p38 lightning split the original activated protein kinase(p-p38),the BCL-2,bax,caspase 3 protein expression changes,and to explore the curative antiapoptotic to dopamine neurons play a role of protection of the specific mechanism of action.Material and method:Select healthy SPF C57BL/6 mice,male,9.5 months old,weight 30 plus or minus 5g,total 60.The mice were randomly divided into normal group,PD model group,medication(Madopar)group,EA group 1 and EA group 2.In addition to the normal group,MPTP(20mg/kg,physiological saline solution),intraperitoneal injection,1/d,and continuous injection of 7d were used to prepare PD mice models.After the models were made,the mice that were not qualified for death and behavioral assessment(climbing bar score < 2 or > 8)were selected,and 8 experimental studies were conducted in each group.Medication(Madopar)group: madopar(50mg/kg,physiological saline solution),total 28d;Normal group and model group:the same amount of physiological saline irrigated stomach,1/d,total 28d;EA group 1 : baihui,shenshu,taixi,100 Hz,continuous wave,20 min,once a day,total 28d;EA group 2: baihui,shenshu,taixi,100 Hz,continuous wave,20 min,once every other day,total 28 d.The behavior of mice was tested by using pole test.Immunohistochemistry tests the number of positive neurons in the black tyrosine hydroxylase(TH)of mice.Western Blot was used to detect the expression levels of TH,p-jnk,p-p38,Bax,bcl-2 and caspase3 in mice of each group,in order to evaluate the related changes of apoptosis.Results: 1.Pole test show: Before modeling,there was no significant difference in the test scores of the mice in each group.After modeling,compared with the normal group,the experimental scores of the mice in other modules were significantly reduced(P<0.01).After the treatment,compared with the model group,the test scores of the mice in the medication group,EA group 1 and EA group 2 were all significantly improved(P <0.01).Compared with the western medicine group,the mice in the EA group 1 had a higher score(P < 0.05).2.TH immunohistochemical staining.The color of TH positive neuron is brownish yellow,mainly distributed in cytoplasm and protuberance.The number of TH positive neurons in the normal group was relatively large,and the color was darker and showed a banded dense distribution.Compared with the normal group,model group mice TH positive neuron cell body and outline is not clear,coloring,remaining neurons form is differ,free nuclei to reduce the bubble,drastically reduced(P< 0.01);Compared with the model group,TH positive neurons of mice in the medicine group,the EA group 1 and the EA group 2 had clear contour,deep dyeing strength and significantly increased number(P < 0.01).Compared with the medicine group,TH positive neurons in the EA group 1 and the EA group 2 were more closely and clearly arranged,with no significant difference in number and no statistical significance(P > 0.05).3.Western blot test results.TH: compared with the normal group,the rest of the module TH protein expression levels were significantly reduced in mice(P < 0.01);compared with model group,the medication group,the EA group 1 and the EA group 2 were significantly higher(P < 0.01);Compared with medication group,the EA group 1 and the EA group 2 of TH protein expression levels were significantly higher(P < 0.01).P-JNK and p-P38 lightning: compared with normal group,the model group,the medication groupand the EA group 1 p-JNK protein expression level was significantly increased(P < 0.01);The rest of the group p-P38 lightning protein expression level significantly increased(P < 0.01).Compared with model group,the EA group 2 p-JNK protein expression level has decreased significantly(P < 0.01);Compared with medication group,including 1 p-P38 lightning protein expression level decreased significantly(P < 0.01).Compared with medication group,the EA group 2 p-JNK expression level decreased significantly(P < 0.01).The bcl-2 and Bax: compared with normal group,model group significantly increased the bcl-2,Bax protein expression level(P < 0.01);the medication group of bcl-2 lower(P < 0.05),Bax significantly reduced(P < 0.01);the bcl-2,Bax protein expression level of the EA group 1 and the EA group 2 significantly reduced(P < 0.01);Compared with model group,the bcl-2,Bax protein expression level of the medication group,the EA group 1 and the EA group 2 decreased significantly(P < 0.01);Compared with medication group,the Bax protein expression level of the EA group 1 decreased significantly(P< 0.01);The bcl-2,Bax protein expression level of the EA group 2 was higher(P < 0.05).Caspase 3: compared with the normal group,the caspase 3 protein expression level of model group was significantly increased(P < 0.01),the medication group,EA group 1 and EA group 2 decreased significantly(P < 0.01);Compared with model group,the caspase 3 protein expression leve lof the medication group,the EA group 1 and the EA group 2 were decreased significantly(P < 0.01);Compared with the medication group,the caspase 3 protein expression level of the EA group 1 and the EA group 2were decreased significantly(P < 0.01).Conclusion: 1."kidney brain of economic" cupping therapy by antiapoptotic role of PD model of substantia nigra dopamine neurons in mice protection,its effect is better than medication Madopar,providing theoretical basis for PD from kidney brain differentiation.2.100 HZ continuous wave electroacupuncture treatment can activate the JNK and p38 signaling pathway through phosphorylation,regulate bcl-2 /Bax ratio through mitochondrial pathway,and finally resist apoptosis by inhibiting the activation expression of caspase-3.3.The electric acupuncture treatment of daily regulation function of every experimental indicators are better than the beauty ba group,to improve the mouse limb movement coordination,improve the substantia nigra TH positive neuron count,p38 lightning protein expression level and the Bcl-2 a day/Bax ratio are superior to treatment.
