Purpose:Observed by experimental study of Hongyuan Pills on bone mineral density in postmenopausal rats with osteoporosis,serum CTX,P1 NP content,bone histomorphometry,bone biomechanics,immunohistochemistry and western blot on TGF-?1 / Smads protein expression situation,explore the Hongyuan pill addition and subtraction of the prevention and treatment of postmenopausal osteoporosis in rats and its mechanism of action,for the red yuan pill addition and subtraction in the prevention and treatment of postmenopausal osteoporosis provide a theoretical basis.Material and methods:80 female rats aged 6 months were selected and divided into normal group,model group(OVX),sham group,high-,medium-,and low-dose Hongyuan pills plus-minus group(HYW)according to the SPSS software.There were a total of 8 groups in the alendronate group(AL)and the GSK(GSK)group.In addition to the normal group and the sham group,all other groups completely removed the bilateral ovaries,and established postmenopausal osteoporosis models.After 6 weeks,the right femur bone mineral density was measured to determine if the model was successful.After successful modeling,the corresponding drug was given to the stomach for 6 weeks.Rats were sacrificed and blood samples were taken to prepare pathological sections.CTX,P1 NP and HE staining were used to measure bone histomorphometry;bone biomechanics was measured;immunohistochemistry and western blot were performed to determine TGF-?1 and Smads proteins in bone tissue.Expression of situation.The data was processed by SPSS 19.0software.Measured data were expressed as mean±standard deviation(x±s).Variance and t-test were used.Count data were expressed in % and x2 test was used.P<0.05 was considered statistically significant.Results:Bone mineral density measurements showed that the bone density of the right femur in the model group was significantly decreased compared with the sham-operated group at 6 weeks after ovariectomy in rats(P<0.01),suggesting a successful model,with high,medium,and low doses of Hongyuan Pills.The addition and subtraction group,alendronate group,and Gushukang group all increased bone mineral density in ovariectomized rats(P<0.05 compared with the model group).The Chinese medicine group had better bone mineral density than the other two traditional Chinese medicine groups(P<0.05),so we used the middle dose Chinese medicine group as a late experimental drug;Elisa staining results showed that compared with the sham group,the CTX in the model group increased and the P1 NP level decreased.(P<0.05),P1 NP increased(P<0.05),CTX levels decreased(P<0.05)in the middle-dose Chinese medicine group compared with the model group;HE staining showed that the percentage of trabecular area decreased in the model group compared with the sham group.The difference was statistically significant(P<0.05).The area percentage of trabecular bone in the medium-dose Chinese medicine group,alendronate sodium group,and the Gushukang group both increased,and there was a statistically significant difference compared with the model group(P<0.05).);bone biomechanical measurement results show that the model The material mechanical properties(elastic modulus)of bone were significantly decreased compared with the sham operation group(P<0.05).The medium-dose Chinese medicine group could improve the material mechanical properties of bone and the difference between the model group was statistically significant(P<0.05);westernblot The results of protein detection and immunohistochemistry showed that the Chinese medicine group,Gushukang group and alendronate group could increase the synthesis of smads protein and TGF-?1 protein in osteoblasts,and there was a statistically significant difference compared with the model group(P< 0.05);The protein was expressed in all groups of rats.The positive density of TGF-?1 and Smads protein in the model group was lower than that in the sham group(P<0.05).The positive density of TGF-?1 and Smads protein in the medium dose group was higher than that in the model group(P<0.05).Conclusion:1.The addition and subtraction of Hongyuan Pills can increase bone mineral density in ovariectomized osteoporosis rats,and have a positive effect on maintaining the three-dimensional spatial structure of trabecular bone and improving osteoporosis in ovariectomized rats.2.The addition and subtraction of Hongyuan Pill can increase the level of serum P1 NP,reduce the level of CTX,and have the effect of promoting bone formation and inhibiting bone resorption.3.The addition and subtraction of Hongyuan Pill can improve the biomechanical properties of bone in ovariectomized rats,increase bone strength,and increase anti-fracture capacity.The addition and subtraction of Hongyuan Pills can increase the expression of TGF-?1protein and Smads protein.This prescription may play a role in preventing and treating osteoporosis in ovariectomized rats by regulating TGF-?1/Smads signal transduction pathway.
