Purposes: 1.To investigate whether the antler polypeptide(VAPs)has a preventive effect on postmenopausal osteoporosis(PMOP).2.To explore the potential therapeutic mechanism of VAPs for the prevention and treatment of PMOP.3.To investigate whether VAPs can regulate the expression of BMP-2/Runx2 protein through the BMP-2/Runx2 signaling pathway to prevent and control PMOP.4.The scientific connotation of the theory of "kidney main bone and essence pulp".Materials and methods:80 female SD rats were randomly divided into 8 groups,10 rats in each group,divided into the normal group(Control),sham operation group(Sham),model group(OVX),high dose group of pilose antler polypeptides(HGAP),dose of pilose antler polypeptides(DGAP)in the low dose group of pilose antler polypeptides(LGAP)and Gushukang group(GSK)and alendronate group(ALN).Take the method to establish the model of ovariectomized osteoporosis after menopause,confirm the model successfully established,intragastric administration started 6 weeks after operation for 6 weeks after injection,the rats by intraperitoneal anesthesia,laparotomy,aorta blood,the bilateral femur and femoral bone density determination;HE staining to observe the trabecular bone micro structure;centrifugal aortic blood serum,the determination of P1 NP,-CTX beta ELISA content;immunohistochemical determination of rat femoral Runx2 level and Western blotting method for the determination of BMP-2 and Runx2 protein expression.Statistical processing: the experimental data were expressed by (?)± S and the data were analyzed with SPSS22.0 software,and the difference was statistically significant with the difference of P< 0.05.Results: 1.Compared with group Control and Sham,the bone density of rats in group OVX decreased significantly,indicating that the PMOP model was established successfully.The high and middle dose group of antler polypeptide could obviously increase the bone density,and the difference was significant,P < 0.05.2.In group OVX,HE staining showed that the structure of trabecular bone was chaotic,the medullary cavity increased,trabecular bone was broken,and the blind end increased.VAPs group could significantly improve the trabecular microstructure,reduce the pulp cavity and reduce the blind end of the fracture,P < 0.05.3.The serum levels of beta-CTX and P1 NP in the control group were significantly higher than those in the normal and sham operated group.The bone metabolism showed a "high conversion" type.The VAPs medium dose group could significantly reduce the level of beta-CTX and P1 NP,and down regulate the bone metabolism level of "high transformation" type.4..Immunohistochemical method and Western blotting method were used to detect the expression of BMP-2 and Runx-2 protein in each group of rats.The results showed that each group of VAPs could significantly improve the expression of BMP-2 and Runx-2 protein,with significant difference(P < 0.05).Conclusions: 1.VAPs can effectively prevent and cure osteoporosis in ovariectomized rats,and the effect of high and middle dose VAPs is better.2.VAPs can prevent PMOP by promoting bone formation,reducing bone turnover rate.3.VAPs may promote osteoblast metabolism by up regulation of the expression of BMP-2 and Runx-2 protein,and prevent postmenopausal osteoporosis.
