Purpose: In this paper,the effects of Pilose antler vessels(PAP)on the proliferation,differentiation and anti-oxidation of bone marrow mesenchymal stem cells(BMSCs)were studied by molecular bioassay technique.Mesenchymal stem cells for extraction,culture and identification.So as to provide a theoretical basis for the better application of antler polypeptide.Method:In this experiment,we observed the morphology of the isolated cells,subcultured and mapped the growth curve,and identified them by the surface antigen markers of BBMSCs.The results showed that the bone marrow mesenchymal stem cells were isolated and cultured.The results showed that the obtained cells were rat BMSCs.The LIF gene was transfected into rat bone marrow mesenchymal stem cells by cell transfection.BMSCs were transfected with LIF.The activity of SOD and MDA was detected by MTT colorimetric assay.And immunohistochemistry were used to observe the effect of antler polypeptide on bone marrow mesenchymal stem cells.Result: In this study,we successfully isolated,cultured and identified rat BMSCs.It was found that the velvet peptide could increase the cell viability,but the cell viability was significantly decreased after transfection of LIF gene,and the velvet peptide could not increase its cell viability,indicating that the antler The effect of the mesenchymal stem cell viability may be through the LIF gene;the antler polypeptide can regulate the antioxidant activity of the cells by regulating the SOD and MDA,and these effects may be achieved by the LIF gene;the antler polypeptide can promote Proliferation and differentiation of rat BMSCs.Conclusion: 1.Velvet peptide can improve the cell viability of bone marrow mesenchymal stem cells;2.Velvet peptides Improve the antioxidant stress activity of bone marrow mesenchymal stem cells;3.velvet peptide can Improve the anti-inflammatory activity of bone marrow mesenchymal stem cells.4.LIF factor transfected into bone marrow mesenchymal stem cells,velvet peptide doesnot affect the proliferation and differentiation of bone marrow mesenchymal stem cells,so consider the transfection of LIF factor bone marrow mesenchymal stem cells as a cell transplant vector. |
