Purpose:To investigate the effects of ShenQi Fuzheng injection on mia-paca-2 cells of human pancreatic carcinoma(proliferation,migration,apoptosis,etc.)and preliminary molecular mechanism studies.Materials and methods: Materials: human pancreatic cancer Mia-PaCa-2 cells and Shenqi Fuzheng injection.Methods:(1)MTT assay was used to observe the effect of ShenQi Fuzheng injection on the proliferation of Mia-PaCa-2 cells in human pancreatic carcinoma with the change of the concentration and time of the dilution drug;(2)The effects of different concentrations of ShenQi Fuzheng injection on the apoptosis of human pancreatic cancer Mia-PaCa-2 cells were detected by flow cytometry and apoptosis detection kit;(3)using Transwell compartment model,To observe the effect of ShenQi Fuzheng injection on the migration of Mia-PaCa-2 cells in human pancreatic cancer;(4)The effects of ShenQi Fuzheng injection on the expression of Bcl-2,ProCaspase-9 and CleavedCaspase-9 protein were detected by Western blot experiment,and the possible molecular mechanism was preliminarily explored.Results: 1.MTT experiment showed that ShenQi Fuzheng injection inhibited the proliferation of human pancreatic carcinoma Mia-PaCa-2cells,showed time concentration dependence,24 h,48h,72 h the highest inhibitory rate was 40%,47%,51%,compared with the same concentration,72 h cell inhibition rate is greater than the 24 h group,Statistically significant,p<0.05.Compared with 48 h experimental group,there was no significant difference in 0.08g/ml and 0.16g/ml drug concentrations after the intervention factor 72 h,p>0.05.2.The flow cytometry detects that ShenQi Fuzheng injection(concentration 0.16g/ml)can promote the apoptosis of Mia-PaCa-2 cells in human pancreatic cancer,and the control group has a statistically significant and p<0.05 in comparison with the early apoptosis rate and late apoptosis rate in the drug group.3.Transwell model experiment shows that ShenQi Fuzheng injection can inhibit the migration of mia-paca-2 cells in human pancreatic cancer,and it has concentration dependence,p<0.05.4.Western Blot experiment showed that the expression of Bcl-2 and ProCaspase-9 protein in human pancreatic carcinoma treated with ShenQi Fuzheng injection was decreased with the increase of drug concentration,p<0.05;but the expression of CleavedCaspase-9 protein showed a tendency to increase with the increase of drug concentration,p>0.05.Conclusions: ShenQi Fuzheng injection can inhibit the proliferation and migration of human pancreatic carcinoma Mia-PaCa-2 cells in vitro;Preliminary mechanism studies indicate that ShenQi Fuzheng injection may reduce the expression of apoptotic protein Bcl-2,ProCaspase-9 and increase CleavedCaspase-9 expression,To promote human pancreatic cancer Mia-PaCa-2 cell apoptosis.It is suggested that ShenQi Fuzheng injection has potential application value in the treatment of pancreatic cancer.
