The Effect Of Xiaochaihu Decoction-containing Serum Combined With Cisplatin On The Apoptosis Of Hep-G2 Liver Cancer Cells

Purpose:To investigate the effects of Xiaochaihu decoction(XCHD)containing drug serum on Cisplatin inducing apoptosis of human hepatocarcinoma Hep-G2 cell,and to verify the mechanism by detecting expression of apoptosis related protein Bcl-2,Bax,and Caspase-3,and to eventually provide laboratory support to clinical use of XCHD combining Cisplatin chemotherapy.Material and method:XCHD solution was prepared based on the recipe from Shang Han Lun.XCHD solution was given to random five rats out of 15 clean rats,three times per day for five days by gavage.The others were given equal amount of saline the same frequency.Blood plasma was collected one hour after the last gavage from abdominal aorta.Hep-G2 cells were divided to three groups: blank serum(BS),blank serum with Cisplatin(BSC),and drug-containing serum with Cisplatin(DCSC).Apoptosis rate of each group was detected by PI method 48 h after giving the cells serum and drugs with three repeats each group.The expression of Bcl-2,Bax,and Caspase-3 of each group was detected by Western blot with three repeats each group.SPSS 21.0 was used to do the statist analysis.Apoptosis was displayed by median,using Kruskal-Hallis H analysis.Protein expression was displayed by average,using multi group single factor ANOVA.?=0.05.Results:1.Median apoptosis rates of three group were DCSC:11.8%,BSC:1.2%,BS:0.6%(p<0.05).2.Bcl-2 expression of DCSC was lower than BSC,and BS was the highest.3.Bax expression of DCSC was higher than BSC,and BS was the lowest.4.Caspase-3 expression of DCSC was higher than BSC,and BS was the lowest.5.Bcl-2/Bax of DCSC was lower than BSC,and BS was the highest.Conclusion:1.Cisplatin can induce Hep-G2 cell apoptosis by affecting mitochondrial apoptosis related protein Bcl-2,Bax and Caspase-3.2.XCHD containing drug serum can enhance the effect of cisplatin promoting Hep-G2 cell apoptosis by affecting Bcl-2,Bax and Caspase-3.3.The results of this research provide laboratory evidence for clinical application of XCHD combining cisplatin chemotherapy in order to enhance the effect of cisplatin and to control the dosage and toxicity of cisplatin.
