The Effect Of Serum Containing Liujunzi Decoction On The Expression Of MMP-9 And Integrin ?1 In A549 Cells Mediated By TGF-?1

Purpose:To observe the effects of serum containing Liu Junzi Decoction on the expressioon of MMP-9 and Integrin?1 in cells A549 mediated by TGF-?1,and to explore the possible mechanism of affecting lung cancer metastasis.Material and method:Preparation the serum containing Liu Junzi Decoction and A549 cells were cu Ltured in vitro.The experiment were divided into the blank serum group,blank serum+TGF-?1 group,and the best containing medicine serum+TGF-?1 group.To observed the intervention degree of TGF-?1 on A549 cells under general microscope.The effect of serum containing Liu Junzi Decoction on TGF-?1 mediated A549 cells migration was observed by scratch test.The expression of MMP-9 and Integrin?1 in protein were detected by immunocytochemical method and Western blot.Resu Lts: 1?TGF-?1 can obviously induce mesenchymal transition of A549 cells and loose connections between cells.2?Compared with blank serum group,the degree of cell migration increased significantly in blank serum+TGF-?1 group(P<0.05);Compared with blank serum+TGF-?1 group,the migration degree of drug serum group decreased(P<0.05).3?Compared with blank serum group,the protein expression of MMP-9 and Integrin?1 increased(P<0.05),Compared with blank serum+TGF-?1 group,the protein expression of MMP-9 and Integrin?1 decreased(P<0.05).Conclusion: 1?The serum containing Liu Junzi Decoction can slow down the migration of A549 cells mediated by TGF-?1.2?The serum containing Liu Junzi Decoction can reduce the expression of MMP-9 and Integrin?1 protein in A549 cells mediated by TGF-?1.3?The serum containing Liu Junzi Decoction can slow down the migration of A549 cells mediated by TGF-?1,which may associated with reduced protein expression of MMP-9 and Integrin?1.
