Effect Of Huangqi Jianzhong Decoction-containing Serum Combined With Cisplatin On The Expression Of E-cadherin And Snail1 In Human Lung Adenocarcinoma A549 Cells

Purpose: To study the effect of Huangqi Jianzhong decoction combined with cisplatin on the expression of E-cadherin and Snail1 in A549 cells of human lung adenocarcinoma.Material and method: A549 cell lines were cultured for 48 h with different concentrations of cisplatin(0,2,4,8,16,32,64ug/ml)in the serum containing Huangqi Jianzhong decoction.Magnetic Tape Terminal was used to detect the inhibition rate of A549 cell lines and determine the optimal concentration of cisplatin.A549 cell lines were divided into four groups: blank group,Chinese medicine group,cisplatin group and combination group.Complete medium,serum containing Huangqi Jianzhong decoction,serum containing cisplatin,serum containing Huangqi Jianzhong decoction and cisplatin were successively used for intervention.After A549 cell lines were cultured for 48 h,the inhibition rate of A549 cell lines in each group was further detected by MTT.The protein expression level of E-cadherin and Snail1 in each group was determined by Western Blot and Immunocytochemistry.The gene expression level of E-cadherin and Snail1 was determined by Realtime-PCR.Results:1.The optimal concentration of cisplatinin combination was 8ug/ml;The inhibition rate of A549 cell lines in the combination group was higher than that in the cisplatin group,the traditional Chinese medicine group and the blank group,the difference was statistically significant(P<0.05).2.In each group of human lung adenocarcinoma A549 cells,the positive expression of E-cadherin was mostly expressed in the cell membrane,while the positive expression of Snail1 was mostly expressed in the cytoplasm,which was stained with brown-yellow granules.The expression of E-cadherin protein in the traditional Chinese medicines group,the cisplatin group and the combination group was higher than that in the blank group.And the expression of E-cadherin protein in the combination group was most.Compared with the blank group,the expression of brown and yellow particles of Snail1 protein in the traditional Chinese medicines group,the cisplatin group and the combination group was less,and the expressionof brown and yellow particles of Snail1 protein in the combination group was least.3.The gene and protein expression level of E-cadherin in the A549 cell lines of the combination group was higher than that in the cisplatin group,the traditional Chinese medicine group and the blank group.The difference was statistically(P<0.05).The gene and protein expression level of the Snail1 in the A549 cell lines of the combination group was lower than that in the cisplatin group,the traditional Chinese medicine group and the blank group.The difference was statistically(P<0.05).Conclusion:Huangqi Jianzhong decoction combined with cisplatin can inhibit the proliferation of A549 cell lines,enhance the expression of E-cadherin protein and gene in A549 cells line of human lung adenocarcinoma,and reduce the expression of Snail1 protein and gene in A549 cells line of human lung adenocarcinoma.
