ObiectiveTo explore the effect of scraping therapy on the prevention and treatment of TUBO breast cancer model mice,and on the prevention of 4T1 breast cancer model mice,and to solve the common problems in scraping research,including appropriate scraping intensity,interval of scraping time,and to explore the immunological molecular mechanism of scraping.MethodsBreast cancer model was established using TUBO breast cancer cells and 4T1 breast cancer cells.24 Balb/c breast cancer model mice were randomly divided into three groups:model group,prevention group and treatment group.The effects of scrapping 24h before and 24h after breast cancer implantation on tumor size in balb/c breast cancer model mice were observed.Another breast cancer model was established with 4T1 breast cancer cells.16 Balb/c breast cancer model mice were randomly divided into two groups:model group and prevention group.The prevention group was scraped 24 hours before breast cancer implantation on tumor size in balb/c breast cancer model mice were observed.The mice were scraped with a quantifiable intelligent scraping plate(1N,3N and 9N),and changes of IL-1? and IL-6 contents in the skin epidermis were observed using HE staining and ELISA.The changes of blood routine at different time points after scraping were detected by whole blood cell analyzer,and the contents of IL-1? and IL-6 in mice at different time points after scrapping were detected by ELISA.T lymphocyte subsets were measured by flow cytometry at different time points after scraping,and the change trend of immune cells and factors at different time points after scraping was preliminarily described.Multiple immune factors(IL-23,IL-1?,IFN-?,TNF-?,MCP-1,IL-12p,IL-1?,IL-10,IL-6,IL-27,IL-17A,IFN-p,GM-CSF,TNF-a,IL-2,IL-17A,IL-4)associated with tumor and inflammation were detected by flow multi-factor test technique at 0.5h,1h and 24h after scrapping.The effect of scraping on the tumor size of mice was investigated by controlling the scraping force with a smart scraping plate.ResultsOn the 15th day after implantation,the breast cancer tissue of mice was removed and weighed.The results showed that the tumor of TUBO and 4T1 breast cancer model mice in scraping prevention group(24h before implantation)decreased,but there was no statistical significance.In the scraping treatment group(24h and 48h after implantation),the TUBO breast cancer decreased,while the 4T1 breast cancer size did not change significantly,and there was no significant difference between the two groups.The results of HE staining showed that there was no significant change in the skin of the 1N scraping group.The skin of the 3N scraping group was not damaged.Blood vessel congestion could be observed but no vascular hemorrhage and inflammatory cell infiltration were observed.The skin of the 9N scraping group was severely damaged,no basic histology was found,the bleeding was serious,and there was more inflammatory cell infiltration.ELISA results showed that the IL-1? content of the 9N group was significantly increased(p<0.01),the IL-6 level was nearly 20 times higher,the difference was significant(p<0.01),and the IL-1? and IL-6 of the 3N group were significantly different(p<0.01).The content was moderately elevated(p<0.05),and there was no significant change in the 1N group.Using the scraping plate controlled the scraping strength,the difference between the TUBO breast cancer prevention group was close to that of the model group,and the tumor size of the prevention group was reduced,but it was not statistically significant.The results of blood routine examination at different time points after scraping showed that the total number of white blood cells and lymphocytes increased gradually from 0.5h to 1h after scraping,decreased at 24h,and returned to normal value after 72h.The results of ELISA at different time points after scraping showed that the levels of IL-1? and IL-6 in serum increased gradually from 0.5h to 1h after scraping,decreased at 24h,and returned to normal level after 72h.The results of flow cytometry detection of T lymphocyte subsets showed that there was no significant change in the percentage of T cells and the percentage of lymphocytes after scraping.The results of flow multi-factor analysis showed that immediately after scraping,IL-10 decreased significantly(p<0.05),IL-10 decreased significantly(p<0.01),IL-6 increased significantly(p<0.01),and MCP-1 decreased significantly(p<0.05)at 0.5 h after scraping.After 24 hours of scraping,MCP-1 returned to normal,IL-10 was still lower than normal(p<0.01),and IL-6 was still higher than normal(p<0.01).ConclusionScrapping may have a preventive effect on the transplanted tumor formed by TUBO and 4T1,and has a certain therapeutic effect on the transplanted tumor formed by TUBO.Preliminary determination of the degree of scraping in mice:3N may be the appropriate intensity of scraping in mice,which can stimulate the immune system and not cause serious damage to the skin tissue.The smart scraping plate can better control the scraping force.Scraping had an effect on the immune system of mice,which was mainly maintained between 0.5h-24h and returned to normal after 72 hours.Scraping can significantly increase the content of MCP-1 and IL-6 in peripheral blood and decrease the content of IL-10,which may be the main mechanism of the effect of scraping on the body.
