Based On Data Mining And Network Pharmacology To Explore The Rules And Mechanism Of Professor Luo Ren’s Medication In The Treatment Of Diabetes

Objective:To explore Professor Luo Ren’s medication rules in the treatment of diabetic nephropathy(DKD),summarize his academic thoughts on the treatment of DKD,and conduct a preliminary analysis of the mechanism of action of his empirical prescription,Qidandihuang Granules.Materials and Methods:1.Collect the case information of DKD patients who meet the inclusion and exclusion criteria in Professor Luo Ren’s outpatient clinic in the past three years,sort out the patient’s gender,age,disease syndrome type,traditional Chinese medicine used in the prescription,and its four qi,five flavors,and meridians.Based on the association rules and cluster analysis of traditional Chinese medicines,Professor Luo Ren’s medication rules for clinical treatment of DKD were obtained.2.Based on Professor Luo Ren’s clinical treatment of DKD’s medication rules,a brief analysis of Professor Luo Ren’s academic thoughts on the treatment of DKD.3.The network pharmacology analysis of Professor Luo Ren’s empirical prescription Qidandihuang Granules was conducted to obtain the key targets of Qidandihuang Granules in the treatment of DKD,and then the enrichment analysis of GO and KEGG pathways was performed on the targets of Qidandihuang Granules in the treatment of DKD.The biological processes,molecular functions,cellular components and signaling pathways mainly involved in the treatment of DKD by Qidandihuang Granules.Result:1.A total of 143 clinical cases and 157 prescriptions were included in this study.Finally,it was concluded that the DKD patients in Professor Luo Ren’s outpatient clinic were mainly qi-yin deficiency syndrome,liver-kidney yin deficiency syndrome,and yin-deficiency dry-heat syndrome.His prescriptions ranked among the top ten The drugs used are yam,salvia,dogwood,licorice,astragalus,fresh rehmannia,rehmannia glutinosa,Moutan cortex,Poria,and Chuanxiong.The medicines used in the prescription are mainly cold in nature,warm in nature,sweet in taste and bitter in taste,and mainly belong to the medicines of the spleen,lung and kidney channels.The analysis of association rules shows strong association medicines such as yam-cornus officinalis,dogwood-rehmannia glutinosa,Moutan cortex-cornus officinalis,salvia miltiorrhiza-astragalus,and salvia miltiorrhiza-fresh rehmannia.Three types of prescriptions are obtained by cluster analysis,and Professor Luo Ren’s treatment of DKD is screened out.The core prescription is Qidandihuang Granules composed of yam,Danshen,Zhigancao,Astragalus,and Rehmannia glutinosa.2.Professor Luo Ren believes that the main pathogenesis of diabetes is deficiency of both qi and yin,and internal knot of anemia.The treatment is based on the principle of nourishing qi,nourishing yin and activating blood.3.Qidandihuang Granules have 161 targets in the treatment of DKD,including 28 key targets,mainly involving 770 biological processes,96 cellular components,160 molecular functions,and 770 signaling pathways.Conclusion:In terms of treatment,Professor Luo Ren discusses the treatment from the perspective of spleen and kidney,combined with the specific conditions of clinical patients,takes spleen and kidney deficiency as the main key point,takes into account blood stasis and diuresis,and conducts syndrome differentiation and treatment in the early,middle and late stages.The core prescription for the treatment of diabetic nephropathy is Qidandihuang Decoction,this prescription has a significant effect on the symptomatic treatment of DKD and delaying the progress of DKD.It can inhibit oxidative stress through multiple targets and multiple channels,reduce renal inflammatory response,reduce cell apoptosis,and delay the process of DKD renal fibrosis.
