Background:Post-stroke depression(PSD)is one of the most common neuropsychological disorders.It is characterized by persistent depression and decreased interest after stroke.At present,the pathogenesis of PSD is not completely clear,and most of them are considered to be the result of a combination of biological,psychological and social factors.In recent years,the changes of intestinal flora associated with neuropsychiatric diseases have received increasing attention.The existing research shows that there is a close relationship between stroke and intestinal microecology.The intestinal flora not only participates in the development of PSD,but also can interfere with PSD.According to the many years of clinical experience of the instructor,the authors put forward the dialectical thinking of kidney deficiency and liver depression in patients with PSD.Combining the characteristics of traditional Chinese medicine and the therapeutic advantages of traditional Chinese medicine,the diversity of intestinal flora in patients with kidney deficiency and liver stagnation type PSD was explored.Objective:In this study,we investigated the diversity of intestinal flora in patients with kidney deficiency and liver stagnation PSD by comparing the differences in intestinal flora diversity between patients with kidney deficiency and liver stagnation and PSD,and explored the mechanism of intestinal flora intervention PSD.Provide new ideas for the treatment of a variety of neuropsychiatric diseases,including PSD.Method:The fecal samples were extracted from 33 cases of post-stroke depression patients with kidney deficiency and stagnation of liver and 10 healthy volunteers,and the DNA of the 16S rDNA V3-V4 region was amplified by polymerase chain reaction(PCR).High-throughput sequencing through the Illumina MiSeqPE platform provides bioinformatics analysis of the final PCR amplification products.Bioinformatics analysis can be used to obtain information on colony composition,species abundance,and community comparison.Results:Compared with the control group,Kidney liver depression patients with PSD group compared with the control group of volunteers of the intestinal flora of alpha diversity analysis showed the ACE,Chao1,Shannon index comparison difference has statistical significance(P<0.05)and control group in ACE,Chaol,Shannon index is higher than kidney liver depression type of PSD group,suggesting kidney liver depression type PSD intestinal flora diversity and abundance of intestinal flora was lower than those of control group.The intestinal flora of PSD patients with kidney deficiency and liver stagnation type was at the phylum classification level,and the relative abundance at the phylum level was higher in the PSD group,while the relative abundance of firmicutes and cyanobacteria was lower and the difference was statistically significant(P<0.05).At eye level in the two groups to compare with bacillus yard,deformation bacteria,lactobacillus high relative abundance in the PSD group,the difference was statistically significant(P<005),YS2)bacterium mesh in the relative abundance of the PSD group is low and the difference was statistically significant(P<0.05)in the relative abundance of the genus level comarison,PSD group of lactic acid bacteria genera,klebsiella bacteria abundance increase,boloodthirsty species,ruminant aureus,MAO spirillum,light steel bacteria genera and rothia,relative abundance of Pasteur's species and was statistically significant(P<0.05).At the phylum level,cyanobacteria and verrucosporium were positively correlated with the HAMD scale score,verrucosporium and age were positively correlated,verrucosporium and proteobacteria were positively correlated with the years of stroke onset,and cyanobacteria and years of stroke onset were negatively correlated,with statistically significant differences(P<0.05).At eye level,bending bacteria mesh,mesh,YS2 bacteria germs warts and HAMD rating scale were positively correlated,lactobacillus yard,warts,micro bacteria were positively correlated with age,yard,Pasteur's bacteria burkholderia bacteria,and Clostridium and negatively correlated with age,yard,intestinal bacteria and germs warts stroke onset age were positively correlated,YS2 bacteria yard,Pasteur's bacteria and negatively correlated with fixed number of year of the incidence of stroke,the difference had statistical significance(P<0.05).On genus level,oxalic acid bacillus genera,campylobacter,ackerman and HAMD rating scale were positively correlated,the Laplace ROM bacteria is a negative correlation with HAMD rating scale,Mr Ackerrmann,optimal coli,dung spore bacteria genera,Mr Gerth bacteria genera,pseudomonas,Anaerotruncus bacteria genera,lactobacillus,coli was positively related with age,fusobacterium,responding to Mr Wolfowitz bacteria genera,Sartre's bacteria genera,bloodthirsty bacteria,bacillus of roses is a negative correlation with age,Pyramidobacter bacteria genera,Mr Ackermann,fusobacterium,dung spore bacteria genera,Mr Gerth bacteria was positively correlated with fixed number of year of the incidence of stroke,Haemophilus and trichotillomonas were positively correlated with the age of stroke onset,and the difference was statistically significant(P<0.05).Conclusions:Intestinal flora of PSD patients with kidney deficiency and liver stagnation was different from that of healthy control group.Chances in intestinal flora abundance in patients with kidney deficiency and liver stagnation type PSD are closely related to various factors of post-stroke depression.To explore the pathogenesis of PSD from the perspective of intestinal flora and to intervene the treat:ment of PSD is worth further exploration.
