Ischemic cerebrovascular disease is a clinically common cerebrovascular disease,accounting for 70%to 80%of all cerebrovascular patients.Cerebral ischemia not only causes damage to brain neurons,but also causes damage to glial cells and microvessels that supply glial cells,neuronal oxygen and nutrients,ie,neurovascular units.Compound Xueshuantong Capsule consists of four traditional Chinese medicines,including Junyao Sanqi,Shengyao Danshen,Zuozhi Yaoqi and Scrophulariaceae.The dosage of Sanqi is about 54.3%.At present,there is no relevant research report on the study of neuroplasticity of compound Xueshuantong after cerebral ischemia-reperfusion injury.In this study,we investigated the effects of compound Xueshuantong Capsule on cerebral ischemia-reperfusion injury in mice by observing neurological dysfunction,neurotrophic factor level and layered neuron morphology and expression of GAP43,Syn,Nogo-A and NgR proteins.The effects of neural plasticity and its related mechanisms.This topic is divided into three parts,and the research contents of each part are summarized as follows:Part Ⅰ:Effect of Compound Xueshuantong on neurological dysfunction in mice with cerebral ischemia-reperfusion inj uryObjective:To prepare the model by middle cerebral artery occlusion(MCAO)method and observe the effect of compound Xueshuantong on neurological dysfunction in mice with cerebral ischemia-reperfusion injury.Methods:Male ICR mice were randomly divided into sham operation group,cerebral ischemia-reperfusion group,compound thrombus low dose(0.75g/kg),medium dose(1.5g/kg)group,high dose(2.25g/kg).The group and the step-head brain heart(0.55g/kg)group,each group were randomly divided into three observation time points of 1day,7days and 14days,and were independently grouped(n=10).Using the MCAO model,the sham operation group only performed anesthesia and blood vessel separation without ligation and embolization.Administered by gavage once a day.Neurological deficits were observed by neurological function scores;sensory motor function impairment was assessed by tape removal and forelimb strength measurement experiments;mouse health status was observed by monitoring body weight and mortality.Results:The sham-operated mice had normal neurological function.The mice in the cerebral ischemia-reperfusion group had obvious neurological deficits.After treatment with compound thrombus,the neurological scores of the mice were reduced and the neurological impairment was improved.In the cerebral ischemia-reperfusion group,the limbs of the mice showed obvious obstacles.The time of removing the adhesive tape on the affected side was prolonged,and the forelimb pull strength of the mice decreased.After treatment with the compound thrombus,the condition of the mice was improved,and the time for removing the tape was shortened.The strength of the forelimbs has increased.In addition,compound thrombosis can improve body weight and reduce mortality after cerebral ischemia-reperfusion.Among them,the compound thrombus low dose group had the best effect.Conclusion:Compound Xueshuantong can improve the neurological function scores of mice with cerebral ischemia-reperfusion injury,reduce the time of removing sticky sticks,enhance the strength of forelimbs,increase body weight,reduce mortality and improve neurological dysfunction in mice.Part Ⅱ:Effect of Compound Xueshuantong on neurotrophic factor levels in mice with cerebral ischemia-reperfusion injuryObjective:To observe the effect of compound Xueshuantong on the levels of NGF,BDNF and NT-3 in mice with cerebral ischemia-reperfusion injury by MCAO model.Methods:Male cerebral ischemia-reperfusion mice were randomly divided into sham operation group,cerebral ischemia-reperfusion group,compound thrombus low dose(0.75g/kg),medium dose(1.5g/kg)group,high dose(2.25g/kg)group and step-head brain heart pass(0.55g/kg)group,each group was randomly divided into three observation time points:1 day,7 days,14 days,and independently grouped(n=10).The model was prepared by middle cerebral artery occlusion(MCAO).The sham operation group only performed anesthesia and blood vessel separation without ligation and embolization.Administered by gavage once a day.The levels of nerve growth factor(NGF),brain-derived neurotrophic factor(BDNF)and neurotrophin-3(NT-3)in brain tissue were determined by ELISA.The levels of neurotrophic factors after cerebral ischemia-reperfusion injury were observed.Results:Compared with the sham operation group,the levels of NGF,BDNF and NT-3 in the brain tissue of mice with cerebral ischemia-reperfusion increased from 1 day to 14 days after surgery,suggesting that ischemia-reperfusion caused nerve damage.Compared with cerebral ischemia-reperfusion,the levels of NGF,BDNF and NT-3 in the brain tissue of the mice in each group were increased,and the compound thrombus low-dose group was significantly increased(P<0.05).Conclusion:Compound Xueshuantong may improve the nerve damage by up-regulating the levels of NGF,BDNF and NT-3 in mice with cerebral ischemia-reperfusion injury.Part Ⅲ:Effect of C ompound Xueshuantong on the damage of cortical neurons in mice with cerebral ischemia-reperfusion injuryObjective:To observe neuronal damage in the ischemic cortex and the expression of GAP43,Syn,Nogo-A and NgR by Nissl staining and immunohistochemical staining.Methods:Male cerebral ischemia-reperfusion mice were randomly divided into sham operation group,cerebral ischemia-reperfusion group,compound thrombus low dose(0.75g/kg),medium dose(1.5g/kg)group,high dose(2.25g/kg)group and step-head brain heart pass(0.55g/kg)group,each group was randomly divided into three observation time points:1day,7days,14days,and independently grouped(n=10).The model was prepared by middle cerebral artery occlusion(MCAO).The sham operation group only performed anesthesia and blood vessel separation without ligation and embolization.Administered by gavage once a day.Immunohistochemical staining was performed after cardiac infusion in each group of mice.Results:Compared with the sham operation group,the Nissl body staining in the cerebral ischemic area of the cerebral ischemia-reperfusion group became shallower,vacuoles appeared,the arrangement was sparse,the number decreased,the number of GAP43 and Syn decreased,Nogo-A,NgR increase the amount.After treatment with compound thrombus,the color of Nissl body became darker,the vacuole decreased,the arrangement improved,the number increased,the number of GAP43 and Syn increased,and the number of Nogo-A and NgR decreased.Among them,the compound thrombus low dose group had the best effect(P<0.05).Conclusion:Compound Xueshuantong Capsule may promote neuronal plasticity and promote neuroprotection by promoting GAP43,Syn expression and inhibiting the expression of Nogo-A and NgR.
