Effects Of Compound Xueshuantong Capsule On PKC Of Retina And IV-C Of Vascular Basement Membrane In DM Rats

Objective:To establish the model of diabetic rats, investigate the effect of Compound Xueshuantong Capsule on the expression of PKC and Ⅳ-C of vascular basement membrane, filter the optimal dosage of Compound Xueshuantong Capsule to treat DR and discuss the mechanism of protecting Visual Function.Methods:Selected10rats as normal group from202healthy SPF SD rats with half males and half females randomly. The rest192rats were injected into the abdominal cavity with50mg/kg weight STZ buffer of2%after fasting8-12hours. Rats of normal group were injected with equal volume saline.Rats with fasting blood-glucose (FBG)≥16.7mmol/L were divided into DM group after1week. DM group rats were grouped into control group, low, middle, high dosages of Compound Xueshuantong Capsule group, Qiming granules group and Doxium group randomly after feeding4mouthes. Each group were given by gavage with corresponding distilled water or drug. After12weeks, executed rats, gathered the eyebals, detached retina, paraffin-embedded and immumohistochemical stained with S-P method. Investigated retinal microvascular structure with electron microscopy and image analysis system Image-Pro Plus6.0to detect the expression of PKC and Ⅳ-C of vascular basement membran. Investigated the inhibitory effect of Compound Xueshuantong Capsule agaist the occurrence and development of DR in DM rats.Results:After being treated12weeks, FBG of the rats in normal group, control group and Doxium group altered hardly(P>0.05), FBG of the rats in low, middle group of Compound Xueshuantong Capsule and Qiming granules group were much lower(P<0.01), FBG of the rats in high group of Compound Xueshuantong Capsule and Qiming granules group were lower(P<0.01).The mean density of PKC protein expression in rats’retina of normal group, control group, low, middle, high group of Compound Xueshuantong Capsule, Qiming granules group and Doxium group were0.1030±0.0160,0.1852±0.0185,0.1731±0.0145,0.1660±0.0121,0.1304±0.0071,0.1380±0.0123,0.1367±0.0140. Compared with that in normal group, The expression of PKC in rats’retina of control group was much higher(P<0.01).Compared with that in control group, the expression of PKC in rats’retina of high group of Compound Xueshuantong Capsule, Qiming granules group and Doxium group was much lower(P<0.01), The expression of PKC in rats’ retina of middle group of Compound Xueshuantong Capsule was lower (P<0.05).Compared with that in Qiming granules group and Doxium group, the expression of PKC in rats’retina of high group of Compound Xueshuantong Capsule was equal (P>0.05).The mean density of Ⅳ-C of vascular basement membran expression in rats’retina of normal group, model group, low, middle, high group of Compound Xueshuantong Capsule, Qiming granules group and Doxium group were0.0860±0.0205,0.2023±0.0228,0.1597±0.0307,0.1402±0.0288,0.0860±0.0187,0.0976±0.0142,0.1049±0.0298. Compared with that in normal group, The expression of IV-C of vascular basement membran of model group was much higher(P<0.01).Compared with that in control group, The expression of IV-C of vascular basement membran in Compound Xueshuantong Capsule group, Qiming granules group and Doxium group was much lower(P<0.01).Compared with that in Qiming granules group and Doxium group, the expression of IV-C of vascular basement membran in rats’retina of High group of Compound Xueshuantong Capsule was equal (P>0.05)Conclusion:Compound Xueshuantong Capsule to some degree can reduce blood glucose, have the effect of restraining the expression of PKC and IV-C of vascular basement membran, reducing thickness in basal membrane of retinal vessels, and probably have inhibitory effect agaist the occurrence and development of DR, and the high group of Compound Xueshuantong Capsuleand is more obvious.
