| Objective: To investigate the protective effects of propofol derivativecqmu-039on cerebral ischemia-reperfusion injury in SD rats and its effectson gliocyte.Methods: A total of40SD rats were randomly divided into5groups:sham surgery group,cerebral ischemia-reperfusion (IR) model group, low(6mg/kg), medium (12mg/kg) and high (24mg/kg) doses of cqmu-039groups. IR model was generated via the surgery of middle cerebral arteryocclusion (MCAO) for2h and then reperfusion of blood for24h. Shamsurgery was performed in the same way as IR model without implanting theembolus. Three different doses of cqmu-039were injected immediatelyafter the IR surgery was performed. Protective effects of cqmu-039wereevaluated through the neurological behavioral score,2,3,5-triphenyltetrazolium chloride (TTC) staining and expressions ofglial cell derived neurotrophic factor (GDNF) and glial fibrillary acidic protein (GFAP) in the hippocampus.Results: Neuronal injury was significantly improved in cqmu-039groups than in model group. In addition, lower neurological behavioralscores, smaller cerebral infarct size and higher expressions of GDNF inCA1, CA3and cortex regions were observed in cqmu-039groups than inmodel group. Expressions of GFAP in CA1, CA3and dentate gyrusregions in cqmu-039groups were decreased in a dose-depended manner.Conclusions: Propofol derivative cqmu-039can effectively improvecerebral ischemia-reperfusion injury of rats through increasing expressionsof GDNF and decreasing expressions of GFAP. |
