Objective: To study the effects of tongduanwan serum on the expression of TLR2,My D88,NF-KB protein in macrophage inflammatory model,in order to provide mechanism for tongduanwan treatment of tubal obstructive infertility,and provide reliable experimental theoretical basis for clinical drug use?METHODS: To establish a SOI model by inoculating female Wistar rats in the vagina with gelatin made of a mixture of Staphylococcus aureus,Mycoplasma Urea and E.coli,and randomly selecting rats to confirm the moulding situation.After the mold was successfully molded,the model group was divided into two groups(experimental group and control group)in a random manner.The experimental group(high dose group of tube pills)and the control group(physiological saline group)were continuously injected for 20 days.After that,After abdominal anesthesia,the rats were collected blood through the abdominal aorta,and the serum was prepared after centrifugation.Then execution,material,naked eye to observe the changes in the tubal morphology of each group;After HE staining,the pathological changes of the tubal tissue of each group were observed under the microscope.The rat peritoneal macrophages were then obtained by primary isolation to establish an inflammatory model of macrophages.The expression of TLR2,My D88,and NF-KB proteins was detected by Western Blot?Result:1.The morphology,color,elasticity and pathological changes of the oviduct tissues in the model group were observed by naked eyes and HE staining,which were consistent with the oviduct inflammatory obstructive changes,indicating that the modeling was basically successful.2.In the macrophage inflammation model,protein expressions of TLR2,My D88 and nf-kb in the tongguan pill group,PPAR-y agonist group and nf-kb blocker group were significantly lower than those in the mixed bacteria group(P<0.05),which may improve the degree of tubal fibrosis and restore the physiological function of the fallopian tube by reducing the release and expression of inflammatory mediators.Conclusion:1.The SOI model can be successfully produced by inoculating gelatin with mixed bacterial solution in the vaginal cavity.2.The protein expressions of TLR2,My D88 and nf-kb in the mixed bacteria group in the macrophage inflammatory model were significantly higher than those in the normal control group(P<0.01),indicating the successful establishment of the inflammatory model.Protein expressions of TLR2,My D88 and nf-kb in the PPAR-y agonist group,nf-kb blocker and tongguan pill group were lower than those in the model group(P<0.05),suggesting that the action mechanism of tongguan pill on SOI is to inhibit the activation of TLR2/My D88/ nf-kb inflammatory signaling pathway by down-regulating the expression of TLR2,My D88 and nf-kb proteins,thereby reducing the inflammatory reaction of the fallopian tubes.
