Objective: By means of aerobic exercise intervention in different stages of depression model rats with 4 weeks of chronic mild and unpredictable stress(CUMS),from the perspective of aerobic exercise on the hypothalamus AVP protein and mRNA in depression model rats,In order to explore the mechanism of exercise affecting depressed rats,it provides a biological theoretical basis for exercise to improve depression from the molecular biology level.Methods: 40 male SD rats with SPF grade of March were selected,the weight 300±20g was randomly divided into 4 groups(N=10): Control group(group C),model group(M Group),Motor Group in Modeling(S1 Group),post-molding exercise group(S2 group).In addition to group C,the remaining three groups were modeled by CUMS method,each group of rats in the mold after the completion of sugar water consumption experiment,absenteeism experiment,whole brain CORT content detection model evaluation,screening out the successful modeling of rats.Among them,the S1 group received 4 weeks CUMS stimulation and aerobic exercise intervention,followed by the experiment of absenteeism and sugar water consumption,after the end of the natural feeding for 4 weeks,S2 group first carried out 4 weeks CUMS stimulation,and then the absenteeism,sugar water consumption experiment,and finally 4 weeks of aerobic exercise.In this experiment,he staining was used to observe the morphological structure of the brain neurons in the hypothalamus of rats,and the expression of AVP was detected by immunohistochemical,Western Blotting and Real-time PCR molecular techniques.Results:(1)Animal Molding Results:(1)General State Observation: group C rats emotional stability,lively and active,fur bright and elastic;M Group,S2 group appeared mood fluctuation,irritable aggressive,eyes without God,diet decreased,hair loss is serious and so on;Compared with group C,the general state of the S1 group was slightly worse;(2)The change of weight growth: before the experiment There was no difference in body weight(P>0.05)in each group,the weight gain of group M,S2 Group and S1 group were significantly lower than that in group C(P<0.01);(3)Total Brain CORT Results: The content of total brain CORT decreased significantly in rats after molding(P<0.01).(2)Behavioral Testing Results:(1)Sugar Water consumption experiment: Compared with group C,the proportion of sugar water consumption in S2 Group and S1 group was significantly lower(P>0.05),and the consumption ratio of sugar water in group M was also significantly lower than that in group C(P<0.01);(2)Absenteeism Experiment: The central area of M Group was lower than group C,S2 Group and S1;The central regional distance of group M is lower than group C,S2 Group(P<0.05);(3)Morphological test Results:(1)HE staining: The neurons in group C rats were evenly distributed and the structure was normal.The neurons in group M were disordered,the cell spacing increased,the number decreased,and the nuclei were fixed and blurred,and the neurons in the S2 group were disordered,the neurons in the S1 group were arranged neatly and the numbers were significantly reduced,and the morphological structure of the neurons in S1 group was significantly worse than that of the S2 group compared with the S2 group.(2)Immunohistochemical Results: Compared with group C,the coloring percentage of AVP positive neurons expressed in the hypothalamus of rats after 4 weeks of CUMS stimulation was higher than that in group C(P<0.01),and the percentage of coloring in M group was significantly higher than that of Group C,S2 Group and S1 Group(P<0.01)compared with M group.(4)Molecular Biology test Results:(1)Real Time-PCR: After modeling,the expression of hypothalamic AVP mRNA in the lower thalamus of M group was higher than that in the remaining three groups,which was statistically significant(P<0.05)(2)Western Blotting: Compared with M Group,the expression of hypothalamus protein in group M was the highest,Very significant higher than group C and S1 Group(P<0.01),and the S2 group is significantly lower than the group M(P<0.05).Conclusion:(1)After 4 weeks of CUMS stress,the Depression model was successfully established;(2)Aerobic exercise can improve the morphological structure of the hypothalamus AVP positive neurons,suggesting that the reduction of AVP expression may be one of the reasons for the occurrence of aerobic exercise to improve depression;(3)In depression,aerobic exercise intervention after depression can regulate the hormones associated with the HPA axis.
