Effects Of Aerobic Exercise On Learning And Memory And Hippocampal SYP、PSD-95 In CUMS Rats

Abstract/Summary:
Objective: At present,depression has become a hot spot in the study of mental diseases at home and abroad,this experiment hopes to find a safer and more effective treatment for it.Methods: In order to investigate the effects of aerobic exercise on learning and memory,SYP and PSD-95 in hippocampus of depressive rats,and to compare the effects of aerobic exercise intervention at different times,we established a CUMS model and performed aerobic exercise interventions at different stages of the model establishment.We selected 48 3-month-old male SPF SD rats as the object of this experiment,they were divided into the following groups:(1)Control group(group C),naturally feed for eight weeks.(2)Model group(group M),after four weeks of establishing CUMS model,naturally feed for four weeks,a total of eight weeks.(3)Model exercise group(group S1)was subjected to four-week aerobic exercise intervention while establishing CUMS model,and then naturally raise for four weeks,a total of eight weeks.(4)Exercise group(group S2),after four weeks of establishing CUMS model,four weeks of aerobic exercise intervention was performed,a total of eight weeks.The body weight of all rats was weighed daily in the experiment.According to the experimental schedule,the sugar-water preference test,the open field test and Elisa tsst,were used to verify the success of establishing CUMS model.The learning and memory abilities of rats were demonstrated by Morris water maze test.After preparing the specimens,we used immunohistochemistry,Western Blotting and Real-Time PCR to detect the expression,protein content and mRNA expression of SYP and PSD-95 in hippocampus of each group.Results:(1)The results of establishing CUMS model:(1)general state: the rats in group C were stable and friendly,and the hair was smooth and elastic,the intake of food and water were basically unchanged;Rats in group M showed anger,irritability,etc.,occasionally there was a fight,the food intake decreased,and the body was thin;The overall situation of rats in S1 group was relatively stable;After exercise intervention in the S2 group,the food intake remained basically unchanged,and the mood tends to be stable.(2)Weighting: Before establishing CUMS model,there was no difference in body weight between the groups.After establishing CUMS model,the body weight of the group M,group S1 and group S2 was lower than that in group C on the 14 th,21st and 28 th day(P<0.01);(3)Sugar-water experiment: After establishing CUMS model,the ratio of sugar-water consumption in group M and group S2 was significantly lower than that in group C(P<0.01),while the percentage of sugar-water consumption in group S1 was higher than that in group M(P< 0.01).(4)Open field test: In group M,the activity,average speed,central area residence time,middle area distance and total distance of rats were significantly decreased(P<0.05).(2)Morris water maze: Localized navigation experiment showed that the average escape latency of rats in each group was shortened and tended to be stable from the third day,and there were significant differences among the groups(P<0.05).In the space exploration experiment,the number of times of rats crossing the original platform in group M decreased significantly(P<0.01).(3)Results of in vitro experiments:(1)Elisa test results: the DA content in the whole brain of M group was significantly lower than that of the other three groups(P<0.01).(2)HE staining: the morphological structure of neurons in hippocampus DG of group C was the most neat;The arrangement of neurons in group M was disordered,the number of cells was significantly reduced,the distance between cells was increased,and the nucleus appeared to shrink and the cell structure was incomplete;The state of hippocampal neurons in S1 group and S2 group was slightly better than that in group M.(3)Immunofluorescence: The IOD values of SYP and PSD-95 in hippocampus DG of group M were lower than those of the other three groups(P<0.05).(4)Western Blotting: The protein content of SYP and PSD-95 in hippocampus of group M was significantly lower than that of the other three groups(P<0.01).(5)Real-time PCR: The expression levels of SYP and PSD-95 in the hippocampus of group M were significantly lower than those of the other three groups(P<0.05).Conclusion:(1)Through the body weight,sugar water consumption experiment,open field test,we found that the behavior of rats was abnormal.Elisa experiment detected the decrease of DA content in the whole brain of rats.These results proved the success of establishing CUMS model from behavioral level and physiological and biochemical leve.(2)The learning and memory abilities of CUMS model were weakened,and the expressions of SYP and PSD-95 in hippocampus were decreased.(3)Aerobic exercise intervention at different stages of establishing CUMS model can increase the expression of SYP and PSD-95 in hippocampus of rats,and then prevent the impairment of spatial learning and memory in time.
