In this work,Rhus chinensis Mill.fruits as the raw materials were extracted by different solvents and the free,esterified,and insoluble-bound phenolics of Rhus chinensis Mill.fruits were also investigated.And aimed to investigate the phenolic composition,antioxidant and pancreatic lipase inhibitory activities of thoes extracts,besides that the effect of thoes extracts on the HepG2 cells induced by H2O2 and the differentiation of 3T3-L1 MBX preadipocytes were also examined.The results were as follows:?1?During the three different solvent extracts,the 80%ethanol extracts and the80%acetone extracts had the high total phenolic contents,and a total of 14 types were identified and quantified by UHPLC-ESI-HRMS/MS.However,as to the food and health industry,it'd better to extract with 80%ethanol.During the three phenolic fractions,the free phenolic fraction had the highest total phenolic content compared with other two fractions and A total of 15 were identified and quantified.Among the 6different extracts,the myricitrin and quercitrin were the main phenolic compounds in Rhus chinensis Mill.fruits.?2?The three different solvent extracts of Rhus chinensis Mill.fruits had a good DPPH·,ABTS+·radical scavenging abilities and there were no significant difference among them.Radical scavenging ratio could reach over 50%,when the concentration were at 2.5?g/m L,besides that in the H2O2 induced HepG2 model,the content of ROS could be reduced with the three different solvent extracts and also reduce the apoptosis.The free phenolic fraction of Rhus chinensis Mill.fruits had the best DPPH·,ABTS+·radical scavenging abilities,compared with other two fractions.In H2O2 induced HepG2 model,the three phenolic fraction had the same trend,which the free phenolic could reduce the ROS contents and the apoptosis sharply than that of two fractions.?3?The three different solvent extracts of Rhus chinensis Mill.fruits had a strong pancreatic lipase inhibitory activities and there were no significant difference among them.However,during the three phenolic fractions,the free phenolic fraction had the best pancreatic lipase inhibitory activities.The pancreatic lipase inhibitory activities of the two main phenolics were also investigated with CalcuSyn calculation and molecular docking.The results suggested that a synergistic effect on the pancreatic lipase inhibitory activities occurred when the concentration of the two compounds were relative high and the binding with the catalytic triad,the binding affinities,hydrogen bonds were possibly involved in the mechanism of the pancreatic lipase inhibitory activities.?4?When the concentration of three different solvent extracts of Rhus chinensis Mill.fruits and the free phenolic fraction were at 60?g/mL,thoes extracts had a good inhibitory effect on the differentiation of 3T3-L1 MBX preadipocytes,and the two main phenolic compounds?myricitrin and quercitrin?and insoluble-bound phenolic fraction had a relative low effect.However,the esterified phenolic fraction didn't have the inhibitory effect on the differentiation of 3T3-L1 MBX preadipocytes.
