Study On The Effect Of Taohong Siwu Decoction On Notch3 Receptor And VEC Proliferation In Rat Vascular Anastomosis Model

Objective: The rat tail vascular anastomosis model as the research object,with Taohongsiwu Decoction intervention group,observation of Taohong Siwu Deco ction on rat tail vascular anastomosis model Notch3 receptor and vascular endo thelial cells(vascular endothelial cell,VEC)proliferation mechanism and interp retation of Taohong Siwu Decoction to promote vascular anastomosis of rat tail model of angiogenesis.Methods: All 40 SPF grade rats were adaptively fed f or 1 weeks.They were randomly divided into 4 groups,namely,peach red Si wu Decoction group,normal saline group,sham operation group and blank con trol group,10 rats in each group.In Taohong Siwu Decoction group and salin e group model,sham group after open exposure vascular suture,blank control group without treatment,and second after operation,two time of 5 days,from each group were randomly selected 5 rats were sacrificed by spinal dislocatio n method,gross observation of anastomosis region the wound,then cut the vas cular anastomosis of blood vessels in the area,the use of immunohistochemistr y to detect the average optical density of Notch3 expression and immunohistoc hemical observation,and rat peripheral blood mononuclear cells were isolated a nd obtained,were cultured in vitro,and repeatedly after purification,detection of VEC proliferation by MTT method.Results: Peach Red Siwu Decoction vascular anastomosis and expression of No tch3 protein was significantly higher than that of saline group,sham operation group and blank control group(P < 0.05),the distinction was striking sense;Taohongsiwu Decoction group the proliferation of VEC stars than in the saline group,sham operation group and blank control group(P < 0.05),the distincti on was striking sense.conclusion:1.Tao Hong Siwu Decoction can increase the expression of angiogenesis relate d factor Notch3 receptor in vascular anastomosis area of rat vascular anastomosis model,thus promoting angiogenesis.2.Tao Hong Siwu Decoction can promote the proliferation of vascular endothe lial cells in the vascular anastomosis area of rats,thus promoting the survival of blood vessels.
