| ObjectiveThis study tried to observe the effects of electroacupunture(EA)"Hegu" point,"Tianshu"point and "Zusanli" point on the stool consistency,visceral sensitivity emotional behavior,and the expression of the change of 5-HT3AR in colon tissues,PKMζ in thalamus of the irritable bowel syndrome(IBS)rat model.And to combine the change of characteristic of neuron discharge in the region of VPL of the brain.To discuss the treatment effects and differences of points on different Meridians,different parts,and different nerve segment to treat the same disease.To illustrates the exist of Specificity of Therapeutic Effects of Acupoints and its relativity,and analyze the rele-vant mechanism in the central and the colon through electro-acupuncture on LI4,ST25 and ST36.MethodsThe neonatal Wistar rats were randomized into the blank control group,the model control group,the "Hegu" group,the "Tianshu" group and the "Zusanli" group.Thirteen rats per group.The other three groups except blank control group were modeled by maternal separation,Acetie acid enema,Combine eolorectal distension(CRD)test.The "Hegu" group,the"Tian-shu" group and the "Zusanli" group aged 2months were given EA,20 minutes every other day for five times,after the IBS models established successfully.The bristol stool score,abdominal withdraw reflection(AWR),elevated plus-maze(EPM)test were observed of rats in each groups.The level of 5-HT3AR in colon tissues,PKMζ in Thalamus were measured wit-h immunohistochemistry and real-time PCR.To observe the changes of VPL neurons by multi-channel in vivo recording technique.Results1.The different effect of the IBS rats’ stool property on different acu-point.Before EA:compared with the blank control group,the Bristol stool score of the model control group,the "Hegu" group,the "Tianshu" group and the "Zusanli" group was increased significantly(P<0.01);Compared with the model control group,There is no signifycant difference of EA groups.Compared with the "Hegu" group,the Bristol stool score of the "Tianshu" group were increased(P<0.05),Compared with the "Tianshu" group,the"Zusanli" group Score reduction(P<0.05).After acupuncture:compared with the blank control group,the Bristol stool score of the model control group and the "Hegu" group,was increased(P<0.01,P<0.05).Compared with the model control group,there is significant difference of EA groups.Compared with the "Hegu" group,the score of the "Tianshu" group,the "Zusanli"group were decreased(P<0.05).Compared with the "Tianshu" group,the "Zusanli" group Score rised;(P<0.05).Before and after acupuncture:EA groups were decreased(P<0.01).2.The different effect of the IBS rats’ intestine sensitivity on different acu-point.Compared with the blank control group,the value of belly raising of the model control group(P<0.01),the "Tianshu" group(P<0.01)and the "Zusanli" gro-up(P<0.01)decreased,and contraction wave increased obviously(P<0.01).Compa-red with the model control group,all of the EA groups of the value of belly raising increased(P<0.01),and contraction wave decreased obviously(P<0.01).3.the influence of the emotional changes at different acupuncture points on IBS modelContrast with the blank control group,the OE%is fewer on the model control group(P<0.01),the "Hegu" group(P<0.01),and the "Tianshu" group(P<0.01),and the "Zusanli"group(P<0.05).the OT%is fewer on the model control group and the "Tianshu" group(P<0.01).Co-mpared with the model control group,the OE%is soared on the "Hegu" group(P<0.05),the "Tianshu" group(P<0.05)and the "Zusanli" group(P<0.01).the OT%increased on the "Hegu" group(P<0.01),the "Tianshu" group(P<0.05)and the "Zusanli"group(P<0.01).Compared with the "Hegu" group,the OE%is raised of the "Zusanli" group(P<0.01).the OT%is fewer on the "Tianshu" group(P<0.05).Compared with the "Tianshu"group,the increased of OE%(P<0.01),OT%(P<0.05)of the "Zusanli" group.4.the influence of the 5-HT3 A receptor expression at different acupuncture pointsContrast with the blank control group,5-HT3A receptor expression increased of the model control group(P<0.01),the "Hegu" group(P<0.01),the "Tianshu" group(P<0.01),and the"Zusa-nli" group(P<0.05).Contrast with the model control group,5-HT3A receptor expression decreased observably(P<0.01)of all EA groups.Contrast with the "Hegu" group,the 5-HT3 A receptor expression decreased of the "Zusanli" group(P<0.01).Contrast with the"Tianshu" group,the 5-HT3 A receptor expression decreased of the "Zusanli" group(P<0.01).5.the influence of the PKMζ mRNA expression at different acupuncture pointsContrast with the blank control group,PKMC mRNA expression decreased of the model control group,the "Tianshu" group(P<0.01),and the "Zusanli" group(P<0.05).Contrast with the model control group,PKMC mRNA expression increased observably(P<0.05)of vHegu" group.Contrast with the "Hegu" grouo,PKMζ mRNA expression decreased of the"Tianshu" group and the "Zusanli" group(P<0.05).6.the influence of the VPL neuron discharge at different acupuncture points on IBS ratUnit Ⅰ:Contrast with the blank control group,mean frequency of the neuron discharge of the model control group(P<0.01),the "Tianshu" group(P<0.05),and the "Zusanli" group(PO.05)were increased.Contrast with the model group,frequency decreased of the "Hegu"group(P<0.01),the "Zusanli" group(P<0.01),and the "Tianshu" group(P<0.05).Contrast with the "Hegu" group,frequency of the "Tianshu" group and the "Zusanli" group were increased of(P<0.05).Unit Ⅱ:Contrast with the blank control group,mean frequency of the model control group(P<0.01),the "Tianshu" group(P<0.05)were increased.Contrast with the model group,frequency of decreased of the "Hegu" group,the "Tianshu" group,and the "Zusanli" group(P<0.01).Contrast with the "Tianshu" group,frequency was decreased of the "Zusanli" group(P<0.05).Conclusion1.The manifestation of model rats5 intestine sensitivity of pain reaction and emoti-onalreaction is abnormal Combined stool score(5-7 score),that is accord with the characteristics of IBS,which is to say that separate the rat from their mother,colonic irritation by acetic acid and intestinal stimulation by dilated balloon can make the D-IBS model.2.EA the three points can reduce the visceral sensitivity,improve abnormal emotional psychological change and the gastrointestinal motility disorder of IBS rat model.Comparison of the three points,"Hegu" point was superior to "Tianshu" and "Zusanli" point of the relieve pain."Tianshu" and "Zusanli" points is better than "Hegu" point of the treatment outcome of gastrointestinal motility disorder."Zusanli" is better to overall regulation,and improve abnormal emotional psychological.This illustrates the exist of Specificity of Therapeutic Effects of Acupoints and its relativity,with the acpoints on different Meridians,different sites,and different nerve segment Can treat the same disease.3.The expression of 5-HT3A receptor increased,and the level of thalamic PKMC mRNA decreased of the IBS model.Acupuncture can inhibit the abnormal expression of 5-HT3A receptors,"Zusanli" is the Strongest point.EA "Hegu" point can enhance the level of thalamic PKMζ mRNA.Prompt the level of the 5-HT3 A receptors and PKMζ mRNA on target organ are participate in the regulation of pain and intestinal motility.And provide the relevant evidence for the specificity of acupoint effect on the molecular biological levels.4.EA can refrain the neuron discharge of VPL of IBS rat.Comparison of the three points,"Hegu" point>"Zusanli">"Tianshu" point of the inhibition of the neuron discharge of VPL.Infer that the two units are related to the regulation of visceral pain of IBS rats.This may provides some basis to confirm the exist of the specificity of the central mechanism. |
