Paeonia ludlowii,a species distributed in secondary shrub forest of Red River Valley and forest edge of Yalu Tsangpo River in the altitude of 2900-3200 metres,belongs to the Paeonia section Moutan DC.of the family of Ranunculacea.According the local chorography,the root bark of Paeonia ludlowii has been used to treat many diseases by Tibetans for hundreds of years,such as gynecological diseases,cardiovascular diseases,dermatomycosis.Epidermophyton floccosum is a kind of Epidermophyton filamentous pathogenic fungus.Trichophyton tonsurans is a kind of Trichophyton filamentous pathogenic fungus.Both of them are pathogenic microorganisms mostly causing clinically tinea capitis and tinea corporis.In this paper,95% ethanol extract of root bark of Paeonia ludlowii was dissolved to the water,then extracted by the petroleum ether,chloroform and ethyl acetate.The antifungal activities were screened and the results indicated that the fraction of ethyl acetate possessed the best antifungal activity.Their minmum inhibitory concentration(MIC)of Epidermophyton floccosum and Trichophyton tonsurans were 3.13-6.25 mg/mL and 1.57-3.13 mg/mL,respectively.Their minmum fungicidal concentration(MFC)of Epidermophyton floccosum and Trichophyton tonsurans were 6.25-12.5 mg/mL and 3.13-6.25 mg/mL,respectively.So it was affirmed that the fraction of ethyl acetate was the effective part for antifungal activity.In this study,twenty-three compounds including seven monoterpenes,three monoterpene glycosides,eight phenols,three phenolic glycosides,one alkaloid and one triterpenoid were isolated from the fraction of ethyl acetate using silica gel column chromategraphy,MCI-Gel column chromatography,Sephadex LH-20 column chromatography,HPLC and identified by means of UV,IR,MS,NMR with referring to the references.The four new compounds were named as paeoniflorone A,paeoniflorone B,paeoniflorone C,2-hydroxy-1-(2-hydroxy-4-methoxyphenyl)propan-1-one.Meanwhile nineteen known compounds,benzoylpaeoniflorin,paeoniflorigenone,2-hydroxymethyl-phenol,vanillic alcohol,paeoniflorin,erythro-1-(3-methoxy-4-hydroxy-phenyl)-propan-1,2-diol,methyl isovanillate,methyl gallate,paeonilide,paeoniflorone,hederagenin,galloylpaeoniforin,benzoic acid,5-hydroxy-6-methyl-1H-indole-3-carbaldehyde,10-hydroxyverbenon,2-hydroxymethylphenyl ?-D-glucoside,paeonol,apiopaeonoside,paeonolide were isolated from the root bark of Paeonia ludlowii,which were isolated from Paeonia ludlowii for the first time.The antifungal activities of twelve compounds isolated from the fraction of ethyl acetate were screened.It was showed that paeoniflorigenone,benzoic acid,methyl gallate and paeonol had presented the obvious activities inhibiting the growth of Epidermophyton floccosum and Trichophyton tonsurans.Their minmum inhibitory concentration(MIC)of Epidermophyton floccosum and Trichophyton tonsurans were 0.25 ?g/mL and 0.25 ?g/mL,0.25 ?g/mL and 0.125 ?g/mL,0.25 ?g/mL and 0.125 ?g/mL,0.125 ?g/mL and 0.0625 ?g/mL,respectively.In the paper,antifungal activity constituents from the root bark of Paeonia ludlowii were researched and twelve monomeric compounds were evaluated for antifungal activities.In summary,it was exhibited that the leading compounds isolated from Paeonia ludlowii provided new medicinal resources for the treatment of dermatomycosis induced by Epidermophyton floccosum and Trichophyton tonsurans.
