| Dryopteris fragrans(L.)schot is a deciduous perennial herb of the Dryopteris genus,which grows mainly in the talc slope of the alpine region,the scree of the forest and lava cracks around volcanoes,mainly distributed in Northeast China,North China,Russia,Japan,Korea,Europe and America.According to the local history records in Wu-da-lian-chi,Dryopteris fragrans decoction has been used to treat many diseases by Daur people for hundreds of years,especially for all kinds of skin diseases.Microsporum canis is a kind of Microsporum filamentous pathogenic fungus,parasitizing cats,dogs,rabbits and other animals,which can infect people,leading to tinea capitis.Epidermophyton floccosum,a kind of filamentous pathogenic fungus which belongs to Epidermophyton,can induce tinea corporis,tinea cruris,tinea pedis,tinea unguium,etc.In the present paper,water extract was subjected to Macroporous resin column chromatography eluted with water,30% ethanol,60% ethanol and 95% ethanol.Then the antifungal activities were screened and found that the sub-fraction of 30% ethanol possessed the best bioactivity,with the minmum inhibitory concentration(MIC)Microsporum canis 0.4-0.8 mg/mL,Epidermophyton floccosum 0.8-1.6 mg/m L,the minmum fungicidal concentration(MFC)Microsporum canis: less than 0.8 mg/m L,Epidermophyton floccosum 1.6-3.2 mg/mL.So it was found that fraction of 30% ethanol was the effective part for inhibiting fungi.In this study,28 compounds including four chromones,five flavones,three terpenes,two coumarins,eleven aromatics and three lignans were isolated from the fraction of 30% ethanol using silica gel column chromategraphy,Sephadex LH-20 column chromatography,HPLC,MCI column chromatography,TLC etc and identified by means of UV,IR,MS,NMR and referring to some references.The four new compounds were named as Dryofrachromen A,Dryofrachromenoside A,Dryofrachromenoside B,Dryofragranoside J.Meanwhile 20 of the 24 known compounds,dihydroconiferylalcohol,Isoscopoletin,pinoresinol,salicifoliol,1,3-dihydroxyl-5-propylbenzene,7-Epiloliolide,3β-hydroxy-5α,6α-epoxy-7-megastigmen-9-one,6-hydroxy-4,7-megastigmadiene-3,9-dione(Dehydrovo mirolio),(E)-3-(4-hydroxyphenyl)acryic acid,salicylic acid,caffeic acid,4-Hydroxy-acetophenone,3,4-Dihydroxyacetophenone,eriodictyol,2-ethyl-6-hydroxybenzoic acid,7,9′;7′,9-diepoxylignanspinoresinol-4,4′-di-O-β-D-glucoside,3,5-dimethoxyphenylglucopyranoside,esculetin,isoorientin and isovitexi were isolated from Dryopteris for the first time.The antimicrobial activities of six known compounds isolated from the part of 30% ethanol were screened.It was showed that dihydroconiferylalcohol,esculetin,5,7-dihydroxy-2-hydroxymethylchromone,(E)-3-(4-hydroxyphenyl)acryic acid had presented the obvious activities inhibiting the growth of Microsporum canis and Epidermophyton floccosum.In the paper,antimicrobial activities of fraction of D.fragrans and monomeric compounds were evaluated.In summary,it was exhibited that the leading compounds which provided new medicinal resources for the treatment of skin diseases induced by Microsporum canis and Epidermophyton floccosum.This program was supported by Heilongjiang Province Natural Science Fund. |
