Paeonia albiflora Pall (Shaoyao) is an important ornamental and medicinal plant in China. The root of Paeonia albiflora Pall. is widely used for the treatment of hepatitis, cirrhosis of liver, cerebral thrombosis and mammary abscess. Chemical studies on Paeonia albiflora Pall. have been carried out since 1969, and the presense of monoterpene glucoside, triterpene, tannin, catechin and aromatic acid have been reported. Various pharmacological activities lead us to investigate the constituents of this plant. Therefore, a systematic chemical research on the constituents of Paeonia albiflora was carried out, which resulted in the isolation of 46 compounds. The chemical structures of 46 compounds were identified by spectral methods, including paeonidanin A(1)~*, paeonidanin B (2)~*, paeonidanin C(3)~*, paeonidanin D(4)~*, paeonidanin E (5)~*, paeonidanin F (6)~*, paeonidanin G(7)~*, paeonidanin H(8)~*, paeoniflorin(9), 4-methylpaeoniflorin(10), benzoylpaeoniflorin(11), 4-methyl-benzoylpaeoniflorin(12), galloylpaeoniflorin(13), 4-methylgalloylpaeoniflorin(14), oxybenzoylpaeoniflorin(15), 6’-O-vanillylpaeoniflorin(16), 4-methyldebenzoyl-paeoniflorin(17), oxypaeoniflorin (18), benzoyloxypaeoniflorin (19), mudanpioside E (20), 6’-O-α-glucopyranosylpaeoniflorin (21), lactiflorin (22), paeonivayin(23), 4-methylalbiflorin(24), paeoniflorigenone (25), (1S, 2S, 4R) -trans-2-hydroxy-1,8-cineoleβ-D-glucopyrannoside (26), (3R, 4S)-3-methyl-3, 4-dihydro-5, 6, 7-trihydroxy -4- (3’-methoxy -4’ - hydroxyphenyl)- 1H-[2]-benzopyran-l-one (27)~*, 4-(2-hydroxy-l-methoxypropyl)-2-methoxyphenol (28)~* , trans-(5-hydroxy-2-methoxy-6-methyl-2, 3-dihydrobenzofuran-3-yl) methyl benzoate (29)~*, cis-(5-hydroxyl-2-methoxy-6-methyl-2, 3-dihydrobenzofuran-3-yl) methyl benzoate (30)~*, 5-hydroxy- 6-methyl-lH-indole-3-carbaldehyde (31)~*, (7S, 8S)-3-methoxy-3’, 7-epoxy-8, 4’-oxyneligna-4, 9, 9’-triol (32), (7’S, 8’R)-dihydrodehydrodiconifery alcohol (33), evofolin B (34), (2R, 3R)-4-methoxyl-distylin(35), 1, 2, 3, 4, 6-pentaO-galloyl-β-D-gIucose (36), catechin (37), benzoic acid (38), gallic acid (39), vanillic acid (40), progallin A (41), methyl gallate (42), resacetophenone (43), β-sitosterol (44), sucrose (45), daucosterol (46). Among them 1-8, 27-31 were new compounds, 26 and 32-35 were isolated for the first time from the plants of this genus.The investigation enriched the constituent types of paeonia albifloria and could provide basic materials for further activity screening.The isolated compounds were evaluated for the inhibitory effects on NO production induced by LPS in N9 Microglia. Cell viability in the present experiment was determined by the MTT method to find whether inhibition of NO production was due to cytotoxicity of the test compounds.The root of Paeonia albifloria has a long history as traditional Chinese medicine and its different processed products showed different pharmacological actions. A simple, sensitive and accurate liquid-chromatographic method with UV detection has been applied for simultaneous determination of six main constituents in different processed product (crude drugs, processed with yellow wine, processed with rice vinegar, stir-frying with bran). The result of the analysis showed similar patterns in HPLC, but differences concerning the content of six constituents, which may have correlation to the pharmacological actions of different processed products.
