Semen Cassiae, the seeds of Cassia obtusifolia L. and Cassia tora L., is a traditional Chinese Medicine. It is reported for the medicinal value as antiseptic, diuretic, diarrheal, antioxidant and protecting liver. Semen Cassiae is distributed mainly in Jiang-su,An-hui,Si-chuan etc provinces of China.Semen Cassiae mainly contains anthraquinone,anthraquinone glycosides, naphtha opyrone, naphthopyrone glycosides. But the research for the chemistry of Semen Cassiae mainly focused on small and medium polar components.We had found that the large polar components have been few studied in our previous work. On the other hand, the method of quality control of Semen Cassiae is to measure the hydrolysates of anthraquinone glycosides. "Chinese Pharmacopoeia" (2005 edition) received the methed of content determination by high performance liquid chromatography for the chrysophanol. and for aurantio-obtusin in 2010 edition. The content of chrysophanol is not less than 0.20%, and aurantio-obtusin is not less than 0.080%. This methods can not reflect the quality of Semen Cassiae accurately and truthfully.On the basis, the paper set up the method of simultaneous measuring 4 compounds of anthraquinones glycosides and naphthopyrone glycosides in Semen Cassiae and determined the 10 batches of Semen Cassiae contents of the 4 compounds. The chemical constituents of Semen Cassiae was systematically analysed, related HPLC fingerprint was set up. Based on fingerprint, the differences between the two species of the herbals was further studied.1 The research of chemical constituents and bioactivities of Semen Cassiae in recent years were reviewed. The constituents of anthraquinone glucosides, naphthopyrone glycosidesand fatty acid were literature evidence-based on as the main constituents.219 compounds have been isolated from the seeds of Cassia obtusifolia L.on the basis of modern chromatography methods, and then the structures of 16 compounds were elucidated by spectroscopic analysis. These compounds were 6 anthraquinone,2 naphthopyrone,4anthraquinone glycosides,3 naphthopyrone glycosides,1 naphthalene glycosides. Among them 4 compounds were newly found ones. These compounds were identified as follows:Six anthraquinone:aurantio-obtusin (6), chryso-obtusin (7), emodin (8), obtusinfolin (9), chrysophanol (12), physicon (13).Two naphthopyrone:rubrofusarin (15-1), toralactone (15-2).Four anthraquinone glycosides:Obtusifolin-2-glucoside (10), aurantio-obtusin-6-O-P-D-glucopyranoside (14), Chrysophanol-1-O-β-D-glucopyranosyl-(1â†'3)-O-β-D-glucopyranosyl-(1â†'6)-O-β-D-glucopyranoside (5), Chrysophano1-1-O-β-D-glucopyranosyl-(1â†'3)-O-β-D-glucopyranosyl-(1â†'6)-O-β-D-glucopyranosyl-(1â†'6)-O-β-D-glucopyranoside (1).Three naphthopyrone glycosides:rubrofusarin gentiobioside (11), Rubrofusarin-6-O-p-D-glucopyranosyl-(1â†'3)-O-β-D-glucopyranosy 1-(1â†'6)-O-p-D-glucopyranos yl-(1â†'6)-O-β-D-glucopyranoside (3), Toralactone-9-O-β-D-glucopyranosyl-(1â†'3)-O-β-D-glucopyranosy1-(1â†'6)-O-β-D-glucopyranosyl-(1â†'6)-O-β-D-glucopyranside (4).One naphthalene glycosides:Torachrysone-8-O-β-D-glucopyranosyl-(1â†'3)-O-β-D-glucopyranosyl-(1â†'6)-O-β-D-glucopyranosyl-(1â†'6)-O-β-D-glucopyranoside (2).3 The HPLC method for determining the content of rubrofusarin gentiobioside, aurantio-obtusin-6-O-β-D-glucopyranoside, casside, cassideB in Semen Cassiae was established firstly, and 10 samples were tested. These compounds are specific and have the pharmacological activities. In addition, the content of them are much higher than the ones used by Chinese Pharmacopoeia (2010 edition) in this drug.4 The fingerprint of Semen Cassiae by HPLC was firstly set up,15 peaks in fingerprint were picked, and 12 peaks of them were confirmed by contrasting with authentic simples. At the same time, the common peaks area were obtained and analysed with clustering analysis, and the differences between the two species in fingerprint were found.
