The disease affects people's lives and can even take away human life.It is of great significance to study the molecular biology mechanisms of diseases to improve the quality of human life.At present,a large number of human disease genes have been included in the OMIM database and the Human Gene Mutation Database.The use of model organisms to construct disease models to study the occurrence and development of human diseases is an important research method in disease research.In recent years,a large number of studies have found that microRNAs(miRNAs)play an important role in the regulation of disease gene expression,such as cardiovascular disease,neurodegenerative diseases,and tumorigenesis.In previous disease models,miRNAs were rarely considered for important regulation of disease genes.Therefore,this paper study the regulation of miRNAs on human disease genes in four model organisms:mouse,zebrafish,fruit fly,and nematode.For this purpose,bioinformatics predictions of miRNA-gene targeting pairs in five species of mice,zebrafish,fruit flies,nematodes,and humans were performed in the genome-wide range;By identifying the genes homologous with human disease genes in mice,zebra fish,fruit flies and nematode genomes,and miRNAs homologous with human miRNAs;In the end,the gene targeting of miRNA-disease genes that homologous with human was selected.In addition,based on Linux+Apache+MySQL+PHP,this paper builds a database of fruit fly disease homologous miRNA-gene.The main research results obtained in this paper are as follows:1.we got 73014 pairs of homologous target pairs between human miRNA-disease-gene and mouse miRNA-gene,which contains 846 mouse miRNAs and 2019 mouse genes,homologous with 868 human miRNAs and 2035 Human disease gene,respectively.2.we got 13013 pairs of homologous target pairs between human miRNA-disease-gene and zebrafish miRNA-gene,which contains 238 zebrafish miRNAs and 802 zebrafish genes,homologous with 260 human miRNAs and 828 human disease genes respectively.3.we got 2263 pairs of homologous target pairs between human miRNA-disease-gene and Drosophila miRNA-gene,which contains 88 Drosophila miRNAs and 352 Drosophila genes,homologous with 144 human miRNAs and 401 human disease genes,respectively.4.we got 490 pairs of homologous target pairs between human miRNA-disease-gene and nematode,which contains 64 nematode miRNAs and 139 nematode genes,homologous with 81 human miRNAs and 163 human diseases,respectively.5.The use of Cytoscape software to visualize the target pair of homology between drosophila and human miRNA-disease genes,and based on Linux + Apache + MySQL+ PHP,the homologous miRNA-gene database was established to provide search,browse and download function(www.njnu.edu.cn/dhdd/home.php).For the first time,this paper has studied homologous miRNA-disease gene targeting pairs in various model organisms(mouse,zebrafish,fruit flies,and nematodes)and constructed a Drosophila disease homologous miRNA-gene database.It provides a reference for the use of multiple model organisms to study the regulation of miRNAs in disease genes.
