HSV-2 is a kind of a-herpes virus subfamily simple genus,consisting of core,cortex,capsid and capsule.It can be spread by close contact and sexual contact to infecting externalia and skin lower trunk and causing vaginitis herpetic,neonatal herpes encephalitis,and genital herpes.Or spreading through the placenta or birth canal,it will cause neonatal congenital infection,result in newborn Death,premature birth and neonatal herpes.In recent years,HSV-2 infection in our country has increased significantly.The results showed that HSV-2 infection rate was about 72.9%in women with STD and reproductive tract inflections,and 25.6%of normal women.System antiviral therapy is the main treatment at present,has achieved some results,but these antiviral drugs is difficult to effectively prevent herpes virus infection,control herpes virus latent infection and recurrent infection.Therefore,vaccination of highly effective and safe HSV-2 vaccine is an effective measure to prevent infection.Rapid progress in technology to promote the great development of new vaccines,people urgently need a comprehensive new vaccine preclinical safety evaluation system,so that human beings can determine the safety and efficacy of vaccine components before clinical trials,can determine the clinical safety dosage,can predict adverse reactions,and can observe all the relationships with toxicity,for clinical monitoring and some cases to provide a reference.Preclinical safety assessment as a part of the overall safety assessment of new vaccines,is one of the important aspects of new vaccine development.At present,subunit vaccines are the most promising new vaccines that can induce cellular immunity and humoral immunity.HSV-2 glycoprotein D(gD)can mediate the fusion of viruses and cells,glycoprotein C(gC)is related to immune escape.In recent years,subunit vaccine is considered a promising candidate vaccine protein.This study explored preclinical safety evaluation of gC2,gD1,gD2 three kinds of glycoproteins.The protective effect of the vaccine was investigated intuitively by the immunoprotective test of guinea pigs.Through the cute toxicity test of BALB/c mice and long-term toxicity test of SD rats to recognize the relationship between the side effects of the vaccine and the quantitative.By the safety pharmacology experiment of Beagle dogs to explore the roles of the main physiological system,and safety of the three protein antigens was verified.
