Objective: To construct a new oncolytic herpes simplex virus type II OHSV2-PD-1-multivary(oHSV2-PD-1v for short)expressing PD-1-Multi Vary,and verify its antitumor activity.Methods: The virus oHSV2-PD-1v was constructed by homologous recombination,and the target gene and expression were detected by PCR and Western blotting.The antitumor activities of virus oHSV2-PD-1v and virus oHSV2-GFP in vitro were compared by MTT assay.The antitumor activity of virus oHSV2-PD-1v in vivo was evaluated by establishing CT26 mouse tumor model.Results: A band with the same size as the target gene PD-1v was obtained by PCR amplification with specific primers.Western blotting was used to detect the PD-1v protein expressed by virus oHSV2-PD-1v after infecting cells.Compared with virus oHSV2-m GMCSF,there is no significant difference in killing tumor cells in vitro.In the model of mouse CT26 colorectal cancer,the virus oHSV2-PD-1v showed a better effect of inhibiting tumor growth and prolonging the growth cycle of mice compared with the negative control group.Conclusion: A novel oncolytic herpes simplex virus OHSV2-PD-1v expressing PD-1V has been successfully constructed,and has the effect of treating tumor. |
