Surface Plasmon Resonance(SPR)sensor can be used to detect the refractive index with high sensitivity,by detecting the properties of reflected light.Compared with other bioanalysis sensors,SPR sensor has higher resolution,lower invasiveness and can achieve real-time monitoring.Therefore,SPR biosensor has been playing an increasingly important role in the field of biological detection.Phase-interrogated SPR sensor has higher resolution than other types of SPR sensors.However,the application of phase-interrogated SPR sensor is limited by the complex phase extraction structure.Based on this background,the main research contents are as follows:1.A phase-interrogated SPR sensor with simple optical system is proposed and constructed.The light path emitted from the SPR sensor chip is divided into three optical paths to detect the intensity of p-light,s-light and 45 ° polarization interference light respectively,to calculate the phase.By using this method,we can obtain the phase change of output light without the help of any phase modulator.Finally,a system,with advantages of simple,stable and convenient,has a resolution of the same order as traditional phase-interrogated SPR sensors.2.The low angle resolution of traditional phase-interrogated SPR sensor system limits the resolution of the whole system.Therefore,an angle adjustment device is designed as a parallel prism controlled by a rotating motor to improve the angle resolution.Based on the proposed modulator-free SPR sensor,the angle adjusting adjustment device can improve the angular resolution to 8.33 × 10-4,and improve the resolution of system to 6.47 × 10-7 RIU.3.The proposed modulator-free SPR sensor was used in the interaction detection of bovine IgG and goat-anti-bovine IgG,and the detection resolution of goat-anti-bovine IgG as high as 1.3881 pM is realized.
