As early as the middle of 20 th century in medicine and biology,researchers has began to explore in the field of cell selection,the separation of different types of cells is the key to predict and treat all kinds of diseases.However,the current cell selection methods are generally expensive,low survival rate,poor screening effective.Some of them need to make specific markers and they also have other shortcomings,which can not meet the needs of scientists.Therefore,this paper has carried out the experimental study,which combination of microfluidic chip technology and cell laser driven technology.Firstily,we analyze and select the cell sampling methods on the ship and make the simulation of hydrodynamic focusing on micro channel with the software of COMSOL.In the simulation,we change the wedth of hydrodynamic focusing to have single cell flow by adjusting the driving pressure of the liquid inlet.At the same time,we have analyzed the optical force and make the study of the mode of cell stress.Both of them provide a theoretical support for the laser driven.Secondly,we developed the research of the methods to make microfluidic chips around the high polymer materials of Polydimethylsiloxane(PDMS in short).The research mainly includes: analysis and select the material of the microfluidic chip;design of chip mask according to the experimental needs;explore the production process of spin coating,lithography,development and chip forming mold;achieve the hydrophilic treatment and chip bonding with oxygen plasma bonding process.Finally,we set up the system platform which is needed for the experiment and analyzed the effect of microfluidic chip on the basis of it.Then,the structure design and optimization of micro flow focusing chip was carried out based on simulation results.We also succeed to achieve the hydrodynamic focusing under 45 degree channel structure which it's width is near 8?m according to controll liquid flow rate with micro injection pump.At this time we have 10?L/min rate in sample liquid channel and 60?L/min rate on both sides of the sheath liquid channel.Besides,we have explored a laser with a wavelength of 980 nm in stationary liquid,which it's minimum power to achieve the cells driven is 50 mW.Based on the above research,the experimental system of laser screening based on microfluidic chip is designed in this paper.This system comprehensive hydraulic focusing,fiber laser and a variety of detection methods on the microfluidic chip and it has broad application prospects and high feasibility,especially for the development of the biomedical field.
